#NOPUBLISH 10/7/2023# Uji aktivitas imunomodulator ekstrak serai dapur (cymbopogon citratus) terhadap histopatologi limfoid tikus putih

ABSTRACT:

Sistem imun yang tidak bekerja secara optimal akan rentan terhadap penyakit. Oleh karena itu untuk meningkatkan sistem imun bekerja secara maksimal dibutuhkan imunomodulator.Salah satu tumbuhan yang dapat berfungsi sebagai imunomodulator alami yaitu serai dapur. Pada penelitian sebelumnya diketahui serai dapur mengandung senyawa yang dapat berfungsi sebagai antimikroba, dan penangkal radikal bebas. Penelitian ini bertujuan untuk mengetahui aktivitas imunomodulator ekstrak serai dapur terhadap gambaran histologi dan indeks organ limpa tikus. Penelitian ini merupakan penelitian eksperimental dengan jumlah sampel hewan coba yang digunakan sebanyak 25 ekor yang dibagi dalam 5 kelompok perlakuan diantaranya : kontrol negatif dengan pemberian CMC-Na, kontrol positif dengan pemberian stimuno R, ekstrak serai dengan dosis 50 mg/Kg BB, dosis 100 mg/Kg BB, dan dosis 150 mg/Kg BB secara oral selama 14 hari. Selanjutnya tikus dibedah dengan mengambil organ limpa dan dibuat sediaan histologi menggunakan blok parafin dan pewarnaan hematoksilin eosin. Parameter yang akan diamati yaitu indeks organ limpa dan pengamatan histopatologi limpa tikus yang terdiri dari pengukuran diameter pulpa putih. Data yang diperoleh akan dianalisis statistik menggunakan One Way ANOVA untuk menentukan pengaruh perbedaan diantara perlakuan. Hasil penelitian menunjukkan perlakuan kontrol negatif memiliki hasil terendah diantara perlakuan yang lainnya. Selanjutnya pada perlakuan yang diberikan ekstrak serai dengan dosis bertingkat menunjukkan peningkatan nilai indeks organ limpa. Sedangkan nilai indeks organ tertinggi yaitu terdapat pada kontrol positif yang dilakukan pemberian stimuno R. Pengamatan pengukuran diameter pulpa putih menunjukkan perbedaan yang signifikan antar perlakuan, hal ini dibuktikan dari hasil statistik P<0,05. Sehingga dapat disimpulkan bahwa ekstrak serai memiliki aktivitas sebagai imunomodulator alami.

ABSTRACT:

An immune system that does not work properly may be susceptible to disease. Therefore, to boost the work of the immune system optimally, it needs immunomodulators. One of the plants that can perform the function of a natural immunomodulator is lemongrass. In previous studies, it was found that lemongrass contains compounds that can serve as antimicrobials and free radical scavengers. This study aims to determine the immunomodulatory activity of citronella extract on histology and spleen index in rats. This study was an experimental study with a total of 25 experimental animal samples which were divided into 5 treatment groups including: negative control with CMC-Na administration, positive control with stimuno R administration, lemongrass extract with a dose of 50 mg/Kg BW, 100 mg dose /Kg BW, and a dose of 150 mg/Kg BW orally for 14 days. Furthermore, the rats were dissected by taking the spleen organs and histological preparations were made using paraffin blocks and hematoxylin-eosin staining. The parameters to be observed were the index of the spleen and histopathological observations of the rat spleen which consisted of measuring the diameter of the white pulp. The data obtained will be statistically analyzed using One Way ANOVA to determine the effect of differences between treatments. The findings showed that the negative control treatment had the lowest results among the other treatments. The treatment of lemongrass extract with graded doses showed an increase in the index value of the spleen organ. While the highest organ index value was found in positive controls that has stimuno R administration. Observations on measurements of the diameter of the white pulp showed significant differences between treatments, this was evidenced by the statistical results P<0.05. So it can be concluded that lemongrass extract has a natural immunomodulator activity.
