Pemisahan Steroid dan Triterpenoid Alga Merah (Eucheuma Spinosum) menggunakan Kromatografi Kolom Vasah dengan variasi ukuran kolom

ABSTRAK

Alga merah (Eucheuma spinosum) memiliki metabolit sekunder antara lain flavonoid, triterpenoid, steroid, alkaloid dan asam askorbat. Tujuan penelitian ini adalah untuk mengetahui pengaruh ukuran kolom dalam pemisahan steroid dan triterpenoid agla merah (Eucheuma spinosum) menggunakan kromatografi kolom. Alga merah Eucheuma spinosum dimaserasi dengan metanol kemudian di dihidrolisis dengan HCl 2 N dan dipartisi menggunakan petroleum eter. Fraksi petroleum eter diuji dengan reagen Liberman-Burchard. Fraksi dipisahkan menggunakan kromatografi kolom dengan variasi ukuran diameter kolom 1; 1,5 dan 2 cm menggunakan eluen n-heksana:etil asetat (17:3) dan perbandingan sampel dan silika 1:100. Fraksi tunggal diidentifikasi UV-Vis, FTIR dan LC-MS. Rendemen proses maserasi dan partisi didapatkan sebesar 5,02% dan 23,07%. Fraksi menunjukkan reaksi positif dengan reagen Liberman-Burchard. Pemisahan kolom 1,5 cm menghasilkan 3 noda tunggal. Panjang gelomang ketiga fraksi adalah 207; 204 dan 206,9 nm memiliki gugus fungsi hidroksi dan geminal dimetil. Hasil LC-MS/MS menujukkan senyawa fukosterol, β-sitosterol, stigmasterol, kampesterol dan desmosterol.

ABSTRACT

Red algae (Eucheuma spinosum) has secondary metabolites such as flavonoids, triterpenoids, steroids, alkaloids and ascorbic acid. The purpose of this research was to determine the effect of column size in the separation of steroids and triterpenoids red algae (Eucheuma spinosum) using column chromatography. Red algae (Eucheuma spinosum) was macerated with methanol and then hydrolyzed with HCl 2 N and partitioned using petroleum ether. The petroleum ether fraction was tested with the Liberman-Burchard reagent. Fraction was separated by column chromatography with variation of column diameter size 1; 1,5 and 2 cm using eluen n-hexane: ethyl acetate (17:3) and the sample ratio and silica 1: 100. Single fraction identified UV-Vis, FTIR and LC-MS. The rendement of maceration and partition process was obtained at 5,02% and 23,07%. The fraction showed a positive reaction with the Liberman-Burchard reagent. The separation of 1,5 cm column got 3 single fractions. The maximum wave length of the third fraction is 207; 204 and 206,9 nm have a hydroxy and geminal dimethyl functional group. The results of LC-MS/MS showed fukosterol, β-sitosterol, stigmasterol, cholesterol and desmosterol compounds.
