Abstract
ABSTRAK
Bekatul merupakan salah satu limbah pertanian yang mengandung karbohidrat 40 % dan kadar pati dalam bekatul sekitar 10 %. Kandungan karbohidrat tersebut dapat dimanfaatkan sebagai substrat dalam pembuatan glukosa melalui hidrolisis secara enzimatis menggunakan enzim glukoamilase menjadi salah satu alternatif sebagai upaya mengurangi jumlah limbah pertanian dan memenuhi kebutuhan glukosa. Penelitian ini bertujuan untuk mengetahui pengaruh suhu dan lama sakarifikasi terhadap proses hidrolisis bekatul menjadi glukosa menggunakan enzim glukoamilase.
Produksi glukosa dilakukan secara likuifikasi menggunakan enzim α- amilase dan dilanjutkan dengan sakarifikasi menggunakan enzim glukoamilase. Tahapan sakarifikasi menggunakan metode Rancangan Acak Lengkap (RAL) yang terdiri dari dua variasi yaitu suhu (50, 60 dan 70 ºC) dan lama sakarifikasi (24, 48 dan 72 jam). Glukosa yang dihasilkan ditentukan dengan metode DNS (asam 3,5-dinitrosalisilat) dengan spektrofotometer pada panjang gelombang 530 nm. Data hasil hidrolisis dianalisis menggunakan analisis varians (ANOVA).
Hasil analisis menunjukkan kadar tertinggi pada perlakuan S2L1 (suhu 60 ºC dan lama sakarifikasi 24 jam) sebesar 29,142 mg/mL (yield glukosa sebesar 8,7 %) dan kadar terendah pada perlakuan S1L3 (suhu 50 ºC dan lama sakarifikasi 72 jam) sebesar 19,540 mg/mL (yield glukosa sebesar 5,9 %).
ABSTRACT
Rice bran is one of the agricultural wastes which contain 40 % of carbohydrates and 10 % starch in rice bran. The carbohydrate content can be utilized as a substrate in the manufacture of glucose on hydrolysis process using glucoamylase enzyme is one of alternatives in reducing the amount of agricultural waste and meet the increasing needs of glucose. The study aims to determine the effect of temperature and length of saccharification on hydrolysis process of rice bran into glucose by using glucoamylase.
The glucose production process was done with liquification using α- amylase enzyme and followed by saccharification using glucoamylase enzyme. Stages of saccharification used complete randomized design (CRD) methode consisted of two variations, those were temperature (50, 60 and 70 ° C) and Times of saccharification (24, 48 and 72 hours). The glucose produced was determined by DNS (acid 3,5-dinitrosalisilat) method was spectrophotometer at wavelength of 530 nm. Data of hydrolysis results were analyzed using analysis of variance (ANOVA).
The reseach showed that highest levels S2L1 treatment (temperature of 60 ºC and 24-hour of saccharification) were 29,142 mg/mL (the yield glucose 7,8 %) and the lowest levels on S1L3 treatment (temperature of 50 ºC and 72 hours of saccharification) were 19,540 mg/mL (the yield glucose 5,9 %).
