Abstract
ABSTRAK
Keadaan hiperglikemia pada penderita diabetes mellitus memicu terjadinya stress oksidatif dan dapat mempengaruhi aktivitas enzim antioksidan alami tubuh. Tanaman binahong merupakan salah satu tanaman yang berpotensi sebagai antidiabetes dan diduga mampu menurunkan stress oksidatif akibat radikal bebas. Penelitian ini bertujuan untuk mengidentifikasi golongan senyawa aktif dan kandungan fenolik total dalam ekstrak etanol umbi binahong, serta mengetahui efek terapinya terhadap aktivitas superoksida dismutase (SOD) hati tikus diabetes.
Ekstrak etanol umbi binahong diperoleh dengan maserasi menggunakan pelarut etanol 70 %. Terhadap ekstrak, dilakukan identifikasi fitokimia dan pemisahan tiap golongan senyawa menggunakan Kromatografi Lapis Tipis Analitik (KLTA) dengan beberapa variasi eluen. Kandungan fenolik total ekstrak diuji berdasarkan metode Folin- Ciocalteu. Selanjutnya dilakukan uji antidiabetes terhadap tikus model diabetes hasil induksi aloksan dengan dosis 25, 50, 75 mg/kg BB. Setelah 14 hari, tikus dibedah dan diambil hatinya untuk pengujian aktivitas SOD. Data aktivitas SOD dianalisis dengan ANOVA dan dilanjutkan dengan uji Duncan (α=0,05).
Hasil penelitian ini menunjukkan bahwa ekstrak etanol umbi binahong mengandung senyawa aktif golongan alkaloid, flavonoid, saponin, tannin, dan terpenoid. Kandungan fenolik total ekstrak umbi binahong terukur sebesar 11,3 mg GAE/L. Bersadarkan hasil analisis, aktivitas SOD hati tikus tiap kelompok perlakuan tidak berbeda nyata (p>0,05). Hal ini menunjukkan bahwa ekstrak umbi binahong tidak mampu meredam radikal bebas dalam jaringan hati tikus diabetes secara maksimal.
ABSTRACT
Hyperglicemia condition in diabetes mellitus patients is able to initiate the oxidative stress and influence the activity of natural body antioxidant. Binahong, a potential plant for antidiabetic treatment is presumed to be having efficacy for reducing oxidative stress caused by free radicals. This study aims to identify the active compound groups and determine total phenolic content in the ethanolic extract of Binahong tuber and recognize its treatment effect on the superoxide dismutase (SOD) activity in the diabetic rat’s liver.
The crude extract of binahong tuber was prepared by maceration in 70 persen of ethanol solvent. The extracts obtained were analysed by phytochemical assay and Analitic Thin Layer Chromatography (ATLC) using variation of eluents. The total phenolic content of the extract was measured by Folin-Ciocalteu method. The antidibetic activity was assayed on the diabetic rat model which induced by alloxan using extract treatment dose of 25, 50, and 75 mg/kg WB, respectively. After 14 days, the rat’s liver were collected for superoxide dismutase (SOD) activity assay. The data obtained was analysed using ANOVA and Duncan test (α=0,05).
The result showed that the ethanolic extract of binahong tuber contain group of alkaloids, flavonoids, saponins, tannins and terpenoids. Total phenols content in this extract was measured about 11,3 mg GAE/L. According to the analysis, rat’s liver SOD activity of each treatment group was not significantly different (p>0,05). It may suggested that ethanolic binahong tuber extract was not able to reduce free radical in the liver tissue of diabetic rats.
