Abstract
ABSTRAK
Untuk mengurangi efek rumah kaca serta memanfaatkan 45 % ampas tebu limbah padat pabrik gula yang belum dimanfaatkan, sehingga dilakukan penelitian mengenai energi alternatif dengan produk biometanol melalui proses hidrolisis gugus metoksil asam poligalakturonat pektin ampas tebu menggunakan ekstrak kasar enzim pektin metil esterase (PMEs) hasil sentesis dari Bacillus subtilis.
Proses hidrolisis gugus metoksil asam poligalakturonat pektin ampas tebu dilakukan dengan variasi konsentrasi ekstrak kasar enzim PMEs (0,5 %, 0,75 %, 1 % dan 1,25 %) hasil optimum pada variasi kosentrasi enzim PMEs selanjutnya dilakukan perlakuan variasi lama waktu inkubasi (1, 2, 3, 4, 5, 6 dan 7 hari). Hasil optimum kadar metanol berdasarkan uji konversi berat jenis (BJ) dari kedua variasi selanjutnya dianalisa secara kualitatif menggunakan instrument kromatografi gas (KG) dan analisa statistik rancangan acak lengkap (RAL) untuk variasi kosentrasi ekstrak kasar enzim PMEs dan rancangan acak kelompok (RAK) untuk variasi lama waktu inkubasi.
Kadar optimum metanol beradasarkan uji konversi BJ pada variasi kosentrasi ekstrak kasar enzim PMEs berada pada kadar PMEs 1 % dengan kadar metanol rata-rata 0,8833 %, sedangkan untuk variasi lama inkubasi kadar metanol optimum pada inkubasi 5 hari dengan kadar metanol rata-rata 1,033 %. Hasil analisa kualitatif metanol sampel menggunakan instrumen kromatografi gas dihasilkan senyawa metanol dan etanol. Analisa data statistik pada variasi kosentrasi ekstrak kasar enzim PMEs dihasilkan tidak beda nyata, sedangkan data statistik variasi lama waktu inkubasi dihasilkan beda nyata.
ABSTRACT
To reduce the greenhouse effect and utilize 45% of solid waste bagasse sugar mills that have not been utilized, so do research on alternative energy through a process of hydrolysis products biometanol metoksil acid groups pectin poligalakturonat bagasse using crude extract of the enzyme pectin methyl esterase (PMEs) results sentesis of Bacillus subtilis.
The process hydrolysis group metoksil acid poligalakturonat pectin bagasse done with crude enzyme extract concentration variation of PMEs (0.5%, 0.75%, 1% and 1.25%) results in the variation of optimum concentration of the enzyme treatment is then performed PMEs long incubation time variation (1, 2, 3, 4, 5, 6 and 7 days). Optimum levels of methanol based on the results of the test weight conversion type (BJ) from the two subsequent variations were kwality analysis using gas chromatography instrument (KG) and statistical analysis of completely randomized design (CRD) for the variation of concentration of crude enzyme extract PMEs and randomized block design (RGD) for the variation of the old incubation time.
Optimum levels of methanol conversion test beradasarkan BJ on enzyme variation in concentration of crude extract PMEs PMEs are at levels of 1% with an average methanol content of 0.8833%, the result of variation in incubation time for optimum levels of methanol in the incubation of 5 days with an average grade methanol 1.033%, the results kwality analysis of gas chromatographic identification of methanol and etanol. Analysis of statistical data on the variation of concentration of crude enzyme extract PMEs produced no real difference, while the statistical variation in the length of time of incubation produced a real difference.
