Abstract
ABSTRACT
Extraction, Toxicity Assay and Identification of Active Compounds Group of Red Algae Extract (Eucheuma cottonii) From Tanjung Sumenep Madura was analysed. The extraction of active compound was conducted through maceration method by using methanol as a solvent. Then condensed extract of methanol hydrolyzed with HCl 2 N and neutralized with NaHCO3. The result of hydrolyzed then partitioned using various solvents that were 1-butanol, ethyl acetate, chloroform, petroleum ether and n-hexane. Toxicity test conducted with BSLT method and Artemia Salina Leach larvae. The identification of the active compound group was conducted with addition of reagent and examined qualitatively.
The result showed that the 1-Butanol is the most effective solvent to extract the active compounds of red algae E. cottonii using liquid extraction method. 1- Butanol fraction is the highest activity of toxicity with LC50 70,32 ppm, followed by ethyl acetate fraction; methanol; Petroleum ether (PE), Chloroform and n-hexane LC50 143,43; 194,40; 195,28; 303,66, and 634,97 ppm. The identification result of active compound group indicated that extract of 1-butanol contain steroids, alkaloids, ethyl acetate extract containing triterpenoids, steroids and alkaloids, methanol extract contains flavonoids, steroids and alkaloids, P.E extract contains triterpenoid, chloroform extract contains flavonoids, triterpenoids and alkaloids, while the n- hexane extract containing triterpenoids and alkaloids.

Ekstraksi, uji toksisitas dan identifikasi golongan senyawa aktif ekstrak alga merah Eucheuma cottonii dari Pantai Tanjung Sumenep Madura telah dianalisis. Ekstraksi senyawa aktif menggunakan metode maserasi pelarut metanol. Ekstrak pekat metanol dihidrolisis dengan HCl 2 N dan dinetralkan menggunakan NaHCO3. Hasil hidrolisat dipartisi menggunakan variasi pelarut 1-butanol, etil asetat, kloroform, petroleum eter dan n-heksana. Uji toksisitas menggunakan metode BSLT dan hewan uji Artemia salina Leach. Identifikasi golongan senyawa aktif dilakukan dengan penambahan reagen dan diamati secara kualitatif.
Hasil penelitian menunjukan bahwa 1-butanol merupakan pelarut yang paling baik untuk mengekstrak senyawa aktif alga merah Eucheuma cottonii menggunakan metode partisi. Pelarut 1-butanol memiliki toksisitas paling baik dengan nilai LC50 sebesar 70,32 ppm, diikuti etil asetat; metanol; Petroleum eter; Kloroform dan n- heksana sebesar 143,43; 194,40; 195,28; 303,66; dan 634,97 ppm. Hasil identifikasi golongan senyawa aktif Eucheuma cottonii menunjukan bahwa ekstrak 1-butanol mengandung steroid, alkaloid, ekstrak etil asetat mengandung triterpenoid, steroid, dan alkaloid, ekstrak metanol mengandung flavonoid, steroid dan alkaloid, ekstrak Petroleum eter mengandung triterpenoid, ekstrak kloroform mengandung flavonoid, triterpenoid dan alkaloid, sedangkan ekstrak n-heksana mengandung triterpenoid dan alkaloid.
