Abstract
ABSTRAK
Allah berfirman dalam Al Qur’an surat Al Maaidah ayat 3 bahwa darah adalah salah satu yang diharamkan bagi umat Islam. Isu keberadaan hemoglobin babi pada filter rokok telah membuat masyarakat yang merokok resah. Diperlukan metode yang mudah untuk menganalisa keberadaan hemoglobin babi pada rokok. Tujuan penelitian ini adalah untuk mengetahui pola khas spektra infra merah pada darah babi dan darah sapi, hemoglobin babi dan hemoglobin sapi, serta filter rokok yang menggunakan hemoglobin babi dan sapi, dengan filter tanpa hemoglobin.
Penelitian ini meliputi: preparasi sampel, terdiri dari tiga perlakuan, yaitu pembuatan larutan PBS (Phosphate Buffer Saline) pH 7,4, menghitung kadar air saren, dan pemisahan hemoglobin dari darah. Identifikasi pola khas spektra protein infra merah pada darah babi dan sapi, hemoglobin babi dan sapi, serta filter rokok yang menggunakan hemoglobin babi dan sapi, dengan filter tanpa hemoglobin menggunakan spektroskopi infra merah pada rentang bilangan gelombang 4000-400 cm-1, resolusi 4 cm-1, dan scan 32. Analisis data spektra infra merah menggunakan metode Second Derivative (2D), dengan teknik diferensiasi Fourier. Validasi data hasil second derivative menggunakan Interpolasi Lagrange.
Hasil penelitian menunjukkan bahwa vibrasi molekuler khas hemoglobin babi dan sapi, serta filter rokok yang menggunakan hemoglobin babi dan sapi, dengan filter tanpa hemoglobin muncul pada turunan kedua spektra infra merah pada bilangan gelombang 580-592 cm-1. Pola spektra turunan kedua hemoglobin sapi terdapat satu puncak satu lembah sedangkan untuk hemoglobin babi menunjukkan satu puncak dua lembah. Hasil second derivative di validasi dengan menggunakan interpolasi lagrange, menunjukkan bahwa pola spektra valid. Jenis vibrasi molekuler daerah spektra khas hemoglobin babi dan sapi pada bilangan gelombang 580-592 cm-1 adalah vibrasi tekukan miring cincin-Fe asimetri (600- 500 cm-1), tekukan N-H (695-550 cm-1), dan uluran C-C (850-500 cm-1) yang masing-masing memiliki lingkungan matriks protein yang berbeda.
ABSTRACT
Allah says in the Qur'an surah Al Maaidah paragraph 3 that the blood is one that is forbidden for Muslims. The issue of the presence of pig hemoglobin in cigarette filters has made people who smoke restless. Needed an easy method to analyze the presence of hemoglobin pig on a cigarette. The purpose of this study was to determine the typical patterns of infrared spectra in pig's blood and the blood of cattle, pig hemoglobin and bovine hemoglobin, as well as cigarette filters which use pig and bovine hemoglobin, with a filter without hemoglobin.
The study include: preparation of samples, consisting of three treatments, namely the manufacture of a solution of PBS (Phosphate Buffer Saline) pH 7.4, to calculate water content Saren, and the separation of hemoglobin from the blood. Identify the typical pattern of the infrared spectra of proteins in the blood of pigs and cattle, pig and bovine hemoglobin, as well as cigarette filters which use pig and bovine hemoglobin, with hemoglobin filters without using infrared spectroscopy in wavenumber range 4000-400 cm-1, a resolution of 4 cm -1, and scan 32. Analysis of infrared spectra data using the method of Second Derivative (2D), with differentiation of Fourier techniques. Validation results of the second derivative data using Lagrange interpolation.
The results showed that the molecular vibrations of typical pork and beef hemoglobin, as well as cigarette filters which use pig and bovine hemoglobin, with hemoglobin filters without appearing on the second derivative infrared spectra in the wavenumber 580-592 cm-1. The pattern of the second derivative spectra of bovine hemoglobin there is one peak of the valley while for pig hemoglobin showed a peak of two valleys. Second derivative in the validation results using Lagrange interpolation, the spectra show that the pattern is valid. This type of molecular vibrational spectra typical of the area pork and beef hemoglobin at wave numbers 580-592 cm-1 is tilted ring-bending vibrations of Fe-asymmetry (600-500 cm 1), N-H bending (695-550 cm-1), and helping C-C (850-500 cm 1) that each have different protein matrix environment.
