Abstract
ABSTRACT
Kidney stones are hard mineral crystals that formed in the kidneys or in the ureter. One way of treatment of kidney stone is by giving a compound that has the potential to dissolve it sueh as green betle leaves sample. The purposes of this research was to determine the amount of kidney stones can be dissolved in aquades extrac of the green betle leaf by in vitro, as well as to determine the concentration of calcium dissolved using an atomic absorption spectrophotometer analysis.
This research consist of sample preparation, production of green betle leaves extract aquades by various concentration 2,5 gr/ 200 mL, 5 gr/ 200 mL, 10 gr/ 200 mL, 15 gr/ 200 mL, flavonoid test, and analysis of kidney stones solubility by weighing 100 mg ± 0,4 mg of kidney stones were incubated on 50 mL various aguades extract of green betle leaves for 3 hours and it were shaked every 20 minutes. The undissolved kidney stones were separated by decantation method. It were dried and weight for determining of dissolved kidney stons. Filtrate was destructed by 65% HNO3 and to determine the concentration of dissolved calcium had analyzed by atomic absorption spectrophotometer.
The result from this research showed a growing number of green betle leaves, the more intense the color is generated, the test reagent showed green betle leaves contain flavonoids. Kidney stone dissolved on a variation of green betle leaves 2,5 gr, 5 gr, 10 gr, and 15 gr respectively of 17.45 mg, 10.65 mg, 13.3 mg and 14.8 mg and calcium levels of dissolved is 470.7984 mg/L, 1716.5884 mg/L, 6389.9884 mg/L, 16006.1384 mg/L. Thus more increasing of concentrated green betle leaves extract aquades higher the concentration of dissolved calcium.
ABSTRAK
Batu ginjal adalah mineral kristal keras yang terbentuk di dalam ginjal atau dalam saluran kemih. Salah satu cara pengobatan batu ginjal adalah dengan pemberian senyawa yang mempunyai potensi untuk melarutkannya seperti sampel pada sirih hijau. Tujuan dari penelitian ini adalah untuk mengetahui berat batu ginjal yang terlarut di dalam ektrak akuades daun sirih hijau secara in vitro, serta mengetahui kadar kalsium terlarut yang dianalisis dengan spektrofotometer serapan atom.
Penelitian ini meliputi preparasi sampel, pembuatan ekstrak akuades daun sirih hijau dengan konsentrasi 2,5 gr/ 200 mL, 5 gr/ 200 mL, 10 gr/ 200 mL, 15 gr/ 200 mL, uji senyawa flavonoid, dan analisis kelarutan batu ginjal dengan cara menimbang batu ginjal utuh seberat 100 mg ± 0,4 mg diinkubasi dalam 50 mL ekstrak akuades daun sirih hijau yang bervariasi selama 3 jam dan setiap 20 menit dilakukan pengadukan selama 5 menit. Batu yang tidak larut dipisah dengan cara dekantasi. Batu ginjal tidak larut dikeringkan dan ditimbang untuk mengetahui berat batu ginjal yang terlarut. Filtrate didestruksi dengan HNO3 65 % dan dianalisis dengan spektrofotometer serapan atom untuk mengetahui kadar kalsium yang terlarut.
Hasil penelitian menunjukkan semakin banyak daun sirih hijau maka warnanya semakin pekat, uji reagen menunjukkan daun sirih hijau mengandung flavonoid. Berat batu ginjal yang dapat larut pada variasi daun sirih hijau 2,5 gr/200 mL, 5 gr/ 200 mL, 10 gr/ 200 mL, dan 15 gr/200 mL secara berturut-turut adalah 17,45 mg, 10,65 mg, 13,3 mg dan 14,8 mg dan kadar kalsium terlarut secara berturut-turut adalah 470,7984 mg/L, 1716,5884 mg/L, 6389,9884 mg/L, 16006,1384 mg/L. Jadi semakin pekat ekstrak akuades daun sirih hijau maka semakin besar pula kadar kalsium yang terlarut.
