Abstract
ABSTRAK
Bawang dayak (Eleutherine palmifolia (L.) Merr.) diketahui mengandung senyawa mayor isoliquiritigenin dan oxyresveratrol yang memiliki aktivitas antikanker. Tujuan penelitian ini untuk menganalisis efek tablet bawang dayak dalam memperbaiki histopatologi kolon dan toksisitasnya terhadap hepar mencit BALB/c model colitis-associated colon cancer (CAC). Penelitian dilakukan dengan studi in vivo menggunakan mencit BALB/c dengan induksi azoxymethane (AOM) 10 mg/kgBB pada hari pertama dan dextran sodium sulfate (DSS) 5% per dua minggu. Pelaksanaan penelitian dilakukan selama 20 minggu. Pada minggu ke-6 dilakukan tes kolitis dengan serum amyloid α (SAA) dan fecal occult blood test (FOBT). Pemberian terapi dilakukan setiap minggu dimulai pada minggu ke-7 dengan dosis tablet bawang dayak 0,25 mg/20gBB, 0,50 mg/20gBB, dan 1,00 mg/20gBB serta doxorubicin 0,052 mg/20gBB. Histopatologi kolon yang diamati yaitu panjang kripta dan jumlah sel goblet sedangkan pada histopatologi hepar yaitu sel normal, degenerasi parenkimatosa, degenerasi hidropik, dan nekrosis menggunakan pewarnaan hematoksilin-eosin (HE). Data yang diperoleh dianalisis statistik menggunakan SPSS 24. Hasil penelitian menunjukkan pemberian tablet bawang dayak 0,50 mg/20gBB dan 1,00 mg/20gBB dapat meningkatkan panjang kripta dan jumlah sel goblet kolon secara signifikan (p<0,05). Dosis 0,50 mg/20gBB juga dapat menurunkan skor histopatologi hepar secara signifikan (p<0,05) sehingga dinilai aman untuk digunakan. Kandungan senyawa oxyresveratrol diduga dapat meningkatkan jumlah sel goblet dengan menurunkan stres oksidatif dan meningkatkan panjang kripta dengan memperbaiki rigiditas tight junction. Senyawa isoliquiritigenin dapat menurunkan ekspresi COX-2 pada kanker kolon dan berfungsi sebagai hepatoprotektor.
ABSTRACT
Bawang dayak (Eleutherine palmifolia (L.) Merr.) is known to contain significant compound isoliquiritigenin and oxyresveratrol which have anticancer activities. The purpose of this study is to analyze the effects of Bawang Dayak tablet in repair colonic histopathology and liver toxicity of BALB/c mice colitis-associated colon cancer (CAC) model. This research is in vivo study using BALB/c mice, which induced by 10 mg/ kgBW azoxymethane (AOM) on the first day and 5% dextran sodium sulfate (DSS) every twice week. This research was conducted for 20 weeks. At 6th weeks it has the colitis test with serum amyloid α (SAA) and fecal occult blood test (FOBT). Therapy started at 7th weeks with 0.25 mg/20gBW, 0.50 mg/20gBW, and 1.00 mg/20gBW dose of bawang dayak tablet in every week. Colonic histopathology were observed the crypt length and total goblet cells, while the liver histopathology were observed normal cell, parenchymatous degeneration, hydropic degeneration, and necrotic using hematoxylin-eosin staining (HE). The data were analyzed statistically using SPSS 24. The result showed that dose 0.50 mg/20gBW and 1.00 mg/20gBW of bawang dayak tablet could increase the colon crypt length and goblet cells significantly (p<0.05). The treatment of 0.50 mg/20gBW bawang dayak tablet could decrease the histopathological score of liver significantly (p<0.05) so it is considered safe to use. The oxyresveratrol compounds are thought can increase the goblet cells and reduce oxidative stress and increase crypt length by fixing the rigidity of tight junction. Isoliquiritigenin compound could downregulating the expression of COX-2 in colon cancer, and it has a function as hepatoprotector.
