Abstract
ABSTRAK
Air cucian beras merupakan limbah yang kandungan nutrisinya masih tinggi. Tujuan dari penelitian ini adalah mengetahui manfaat air limbah cucian beras sebagai media alternatif untuk membiakkan bakteri endofit kitinolitik (Pseudomonas pseudomallei, Klebsiella ozaenae) dan Bacillus thuringiensis serta pengaruhnya terhadap mortalitas dan perubahan morfologi nyamuk Aedes aegypti. Penelitian dilakukan dengan menggunakan rancangan percobaan faktorial, dengan kombinasi perlakuan ditempatkan menurut RAL dan diulang 3 kali.
Media pertumbuhan bakteri berupa air limbah dari pencucian 400 gr beras giling dengan 400 ml air, kemudian difiltrasi dengan kertas saring. Viabilitas bakteri dihitung dengan metode TPC. Perlakuan yang diberikan pada larva nyamuk yaitu empat variasi suspensi (B. thuringiensis, P. pseudomallei, K. ozaenae, dan kombinasi antara P. pseudomallei dan K. ozaenae) dengan konsentrasi 0 ml, 0,5 ml, 1 ml, dan 1,5 ml. Masing- masing diberikan untuk 20 ekor larva uji instar III yang dipelihara dalam wadah yang berisi 150 ml air. Pengamatan terhadap mortalitas larva diamati setiap 1x24 jam. Jumlah larva yang mati di analisis dengan ANOVA program SPSS 16. Abnormalitas morfologi larva pada kelompok perlakuan dibandingkan dengan larva normal pada kontrol.
Hasil pengamatan menunjukkan, air limbah cucian beras dapat dimanfaatkan sebagai media alternatif untuk membiakkan bakteri endofit kitinolitik dan B. thuringiensis. Hal ini dibuktikan dengan viabilitas P. pseudomallei pada media ini mencapai 1,85 x 108 cfu/ml, K. ozaenae 3,19 x 108 cfu/ml, dan B. thuringiensis 5,3 x 107 cfu/ml. Suspensi bakteri tersebut berpengaruh terhadap mortalitas larva dan pupa nyamuk Aedes aegypti. Diantara empat macam suspensi bakteri, suspensi B. thuringiensis dengan konsentrasi 1,5 ml merupakan yang paling efektif dalam membunuh larva dan pupa nyamuk A. aegypti, yaitu dengan persentase 100% dalam waktu 5 hari. Hasil penelitian juga menunjukkan bahwa suspensi bakteri endofit kitinolitik dan B. thuringiensis berpengaruh terhadap kerusakan morfologi larva dan pupa nyamuk Aedes aegypti.
ABSTRACT
Rice washing water is waste that is still high nutritional content. The purpose of this study was to determine the benefits of rice wastewater as an alternative medium for culturing endophytic chitinolytic bacteria (Pseudomonas pseudomallei, Klebsiella ozaenae) and Bacillus thuringiensis and its influence on mortality and morphological damaged of Aedes aegypti mosquito. The study was conducted using a factorial experimental design, with the treatment combination were placed according to RAL and 3 repetition.
Cultural media of bacteri are from waste water by washing grist 400 gr with 400 ml of water, then filtered with filter paper. Viability of bacteria calculated by the TPC method. The treatment given to the mosquitoes larvae are four variations of the suspension (B. thuringiensis, P. pseudomallei, K. ozaenae, and combination of P. pseudomallei and K. ozaenae) with concentration 0 ml, 0.5 ml, 1 ml, and 1.5 ml. Those are given to 20 third instar larvae test kept in containers that containing 150 ml of water. Observations of larval mortality was observed every 1x24 hours. The number of dead larvae were analyzed by ANOVA test of SPSS 16 program. Larval morphological abnormalities in the treatment group compared with normal larvae in control.
The results showed, rice wastewater can be used as an alternative media for culturing endophytic chitinolytic bacteria and B. thuringiensis. Evidenced by the viability of P. pseudomallei in this medium reached 1.85 x 108 cfu / ml, K. ozaenae 3.19 x 108 cfu/ml, and B. thuringiensis 5.3 x 107 cfu / ml. The bacterial suspension affect on mortality of Aedes aegypti larvae and pupae. Among the four kinds of bacterial suspension, B. thuringiensis suspension of 1.5 ml was the most effective in killing A. aegypti mosquito larvae and pupae, which is the percentage of 100% within 5 days. Another result showed that suspension of endophytic chitinolytic bacteria and B. thuringiensis had effect on Aedes aegypti larvae and pupae morphological damage.
