Abstract
ABSTRAK
Demam berdarah dengue (DBD) merupakan penyakit yang ditularkan oleh nyamuk Aedes aegypti. Pemberantasan nyamuk A.aegypti umumnya masih mengandalkan penggunaan insektisida kimiwai yang memiliki dampak negatif berupa resistensi serangga, matinya organisme bukan sasaran dan pencemaran lingkungan. Tujuan dari penelitian ini adalah mengetahui pengaruh bakteri endofit kitinolitik (Pseudomonas pseudomallei, Klebsiella ozaenae) dan Bacillus thuringiensis terhadap mortalitas dan perubahan morfologi nyamuk A.aegypti yang ditumbuhkan dan diperbanyak di media air kelapa.
Jenis penelitian ini adalah eksperimental dengan Rancangan Acak Lengkap (RAL) yang terdiri dari dua faktor perlakuan yakni jenis bakteri (bakteri endofit kitinolitik dan B.thuringensis) dan konsentrasi suspensi bakteri yaitu 0 ml sebagai kontrol, 0,5 ml, 1 ml, dan 1,5ml. Masing-masing diberikan untuk 20 ekor larva uji instar III yang dipelihara dalam wadah yang berisi 150 ml air. Media pertumbuhan bakteri berupa air kelapa dan TSB sebagai media kontrol dan pembanding . Air kelapa sebanyak 300ml difiltrasi dengan kertas saring dan disterilisasi dengan autoclave. Pengamatan terhadap mortalitas larva diamati setiap 1x24 jam selama 10 hari. Daya hidup bakteri dihitung dengan metode TPC (Total Plate Count). Jumlah larva yang mati di analisis dengan program SPSS 17 Two Way Anova dan dilanjutkan dengan uji lanjut Duncan. Nilai LC50 dianalisis dengan program Regressi Probit. Mortalitas dan kerusakan morfologi larva dan pupa pada kelompok perlakuan dibandingkan dengan larva dan pupa normal pada kontrol.
Hasil pengamatan menunjukkan, air kelapa dapat dimanfaatkan sebagai media alternatif perbanyakan bakteri endofit kitinolitik (P.pseudomallei dan K. ozaenae) dan B.thuringiensis. Pada media air kelapa, daya hidup bakteri P.pseudomallei sebesar 3,76 x 108 cfu/ml, K. ozaenae sebesar 4,26 x 108 cfu/ml dan B.thuringensis sebesar 4,05 x 108 cfu/ml. Hasil pengamatan juga menunjukkan diantara variasi perlakuan, interaksi jenis bakteri Bacillus thuringensis pada konsentrasi 0,5ml, 1 ml, 1,5 ml dan kombinasi P.pseudomallei dan K.ozaenae pada konsentrasi 1,5ml memiliki pengaruh mortalitas terhadap larva paling tinggi dengan persentase sebesar 100% pada hari kesepuluh pengamatan. Ditemukan nilai LC50 yang efektif mematikan larva sebesar 50% dalam waktu 10 hari pengamatan. Hal ini menunjukkan penggunaan konsentrasi yang lebih tinggi akan mematikan larva A. aegypti dalam jumlah yang banyak.
ABSTRACT
Dengue hemorrhagic fever (DHF) is a disease transmitted by the Aedes aegypti mosquito. Eradication of Aedes aegypti mosquitoes generally still rely on the use of chemical insecticides that have a negative impact in the form of insect resistance , the death of non-target organisms and environmental pollution . The purpose of this study was to determine the effect of chitinolytic endophytic bacteria (Pseudomonas pseudomallei , Klebsiella ozaenae ) and B. thuringiensis on mortality and morphological changes in the mosquito A.aegypti which are grown and propagated in coconut water as the media .
This type of research is experimental research with completely randomized design (RAL), which consists of two types of treatment factors for chitinolytic endophytic and B.thuringensis bacteria, and the concentration of 0 ml bacterial suspension as a control , 0.5 ml , 1 ml , and 1 , 5ml . Each one was given to 20 third instar larvae, and was maintained test in containers containing 150 ml of water. Bacterial growth media is in the form of coconut water and TSB as media control and comparison. 300ml coconut water is filtered with filter paper and sterilized by autoclave. Observations on larval mortality were observed for every hour in 1x24 hour within 10 days. Power is calculated by the method of bacterial life TPC (Total Plate Count). Numbers of dead larvae were analyzed by SPSS 16 Two Way ANOVA’s and Duncan followed with further testing. LC50 values were analyzed using Probit Regressi program. Mortality and damage morphology of larvae and pupae in the treatment group compared with normal larvae and pupae in control.
The results showed, coconut water can be used as an alternative media propagation chitinolytic endophytic bacteria (P.pseudomallei and K.ozaenae) and B. thuringiensis . On coconut water medium , bacteria P.pseudomallei survival of 3.76 x 108 cfu / ml , K.ozaenae of 4.26 x 108 cfu / ml and B.thuringensis 4,05 x 108 cfu / ml . The observation also showed variation between treatments , the interaction of bacteria B.thuringensis at a concentration of 0.5 ml , 1 ml , 1.5 ml , and the combination of P.pseudomallei and K.ozaenae at 1.5 ml concentration has an influence on larval mortality with the highest percentage of 100 % on the tenth day of observation . LC50 value was found effective lethal larvae by 50 % within 10 days of observation. This suggests that the use of a higher concentration will kill the larvae of A.aegypti in large numbers .
