Abstract
ABSTRAK
Keberadaan logam berat di lingkungan perairan sangat berbahaya bagi kelangsungan mahluk hidup didalamnya maupun bagi manusia, khususnya logam berat timbal (Pb) yang paling banyak digunakan dalam perindustrian sehingga limbahnya dapat mencemari lingkungan. Alternatif pengolahan limbah logam berat Pb dengan menggunakan bakteri sebagai biosorben. P. pseudomallei merupakan bakteri gram negatif yang tumbuh di tanah dan air. Tujuan dari penelitian ini adalah mengetahui pengaruh konsentrasi logam berat timbal (Pb) terhadap pertumbuhan P. pseudomallei serta kemampuan P. pseudomallei dalam menurunkan konsentrasi logam berat timbal (Pb).
Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 5 perlakuan dan 3 kali ulangan. Bakteri P. pseudomallei dibiakkan dalam media LB dan dibuat kurva standar pertumbuhan diamati jumlah koloni (TPC) yang diregresikan dengan nilai OD panjang gelombang 660 nm menggunakan persamaan y=ax+b. Pengamatan pertumbuhan bakteri dengan konsentrasi logam berat timbal 0,5,10,15,20 ppm dihitung dengan melihat OD 660 nm setiap 4 jam sampai 28 jam. Untuk mengetahui konsentrasi logam berat timbal yang terserap menggunakan metode Atomic Absorbtion
Spektrofotometry (AAS) diambil dari hasil pertumbuhan optimal selain kontrol.
Hasil pengamatan menunjukkan, bakteri P. pseudomallei berpotensi sebagai biosorben logam berat timbal (Pb). Hal ini dibuktikan dengan pertumbuhan optimal bakteri P.pseudomallei pada konsentrasi logam berat timbal (Pb) 5 ppm dengan nilai OD 1,87 dengan jumlah koloni 1,19x1010cfu/ml. Dan terendah 20 ppm OD 1,56 jumlah koloni 1x1010cfu/ml. Kadar logam berat timbal (Pb) yang terserap oleh bakteri P. pseudomallei yaitu dengan persentase 98,2% pada konsentrasi 5 ppm.
ABSTRACT
The presence of heavy metal are very dangerous for the survival of living things as well as for humans, especially heavy metal lead (Pb) are the most widely used in industry and that waste can pollute the environment. Alternative prosessing of waste heavy metal Pb using bacteria as biosorbent. P. pseudomallei is a gram-negative bacterium that grow in soil and water. The aims of this research is effect of the consentration of heavy metal lead on growth and the ability bacteria P. pseudomallei in reducing heavy metal concentration of lead (Pb).
This research is using completely randomized design (RAL) with 5 treatments and 3 replications. P. pseudomallei bacteria cultured in media LB and made a standard curve method expressing the relation of growth with colony counts (TPC) regressed to the value of OD 660 nm wavelength, using the regression equation y=ax+b. the research of bacteria growth with concentrations of heavy metal lead (0, 5, 10, 15, and 20 ppm) calculated by looking at OD 660 nm every 4 hours to 28 hours. To find out concentrations of heavy metal lead is absorbed using Atomic Absorbtion Spektrofotometry (AAS) is taken from the results of optimal growth than the control.
The results of research showed that the potential of bacteria P. pseudomallei as biosorbent of heavy metal lead (Pb). This showed by the optimal growth of bacteria P. pseudomallei in heavy metal lead 5 ppm with value OD 1,87 and colony counts 1,19x1010cfu/ml and lowest growth at 20 ppm with value OD 1,56 and colony count 1x1010cfu/ml. levels of heavy metal lead (Pb) are absorbed by bacteria P. pseudomallei with the percentage of 98,2% at the concentration 5 ppm.
