Abstract
ABSTRAK
Daun sisik naga (Drymoglossum piloselloides) merupakan salah satu tanaman tradisional yang mengandung senyawa flavonoid. Senyawa ini diketahui mampu menghambat stimulasi StAR yang berfungsi memediasi kolesterol dari sitoplasma menuju membrane mitokondria dan P450 scc yang mengubah kolesterol menjadi pregnenolon. Penelitian ini bertujuan untuk mengetahui efek pemberian ekstrak etanol daun sisik naga (Drymoglossum piloselloides) terhadap berat dan histologi testis tikus putih (Rattus norvegicus).
Penelitian ini bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 6 kali ulangan. Perlakuan yang digunakan adalah K (kontrol), P1 (5,04 mg/200 BB ), P2 (7,56 mg/200 BB), P3 (10,08 mg/200 BB) dan P4 (12,6 mg/BB). Hewan yang digunakan adalah tikus putih jantan strain Wistar sebanyak 30 ekor yang berumur ± 2-3 bulan dengan berat rata-rata 230 gr. Data hasil penelitian meliputi jumlah sel leydig, jumlah sel sertoli, sel germinal (spermatogonium, spermatosit primer, spermatosit skunder dan spermatid), berat testis dan keliling tubulus seminiferus. Data yang diperoleh dianalisis dengan ANOVA One Way, apabila terdapat perbedaan sangat nyata maka dilanjutkan dengan uji lanjut Duncan α 1%.
Hasil penelitian menunjukkan bahwa ekstrak etanol daun sisik naga (Drymoglossum piloselloides) berpengaruh dalam menurunkan, jumlah sel leydig, sel sertoli, sel germinal dan berat testis, sedangkan keliling tubulus seminiferus tidak berpengaruh. Penelitian ini memperlihatkan bahwa ekstrak etanol daun sisik naga (Drymoglossum piloselloides) berpengaruh menurunkan berat testis dengan dosis terbaik P4 (12,6 mg/200g BB) dan jumlah sel leydig, sel sertoli serta sel germinal menurun secara efektif pada dosis P1 (5,04 mg/kg BB). Namun pada keliling tubulus seminiferus tidak terjadi penurunan secara signifikan.
ABSTRACT
Leaf dragon scales ( Drymoglossum piloselloides ) is one of the traditional plants containing flavonoids . These compounds are known to inhibit the functional StAR mediated stimulation of cholesterol from the cytoplasm to the mitochondrial membrane and P450 scc that converts cholesterol to pregnenolone . This study aimed to determine the effect of ethanol extract of leaves of dragon scales ( Drymoglossum piloselloides ) on testis weight and histology of white rats ( Rattus norvegicus ) .
This is an experimental study using a completely randomized design ( CRD ) with 5 treatments and 6 replications. The treatment used is K ( control ), P1 ( 5.04 mg/200 BB ), P2 (7.56 mg/200 BB ), P3 ( 10.08 mg/200 BB ) and P4 ( 12.6 mg / BB .) Animals used were Wistar strain male rats 30 tails ± 2-3 months years old with an average weight of 230 gr. Research data include the number of Leydig cells, the number of Sertoli cells, germ cells ( spermatogonia, primary spermatocytes, secondary spermatocytes and spermatids ), testis weight and circumference of the seminiferous tubules . Data were analyzed with ANOVA One Way , where there is a very real difference then followed by a further test Duncan α 1 % .
The results showed that the ethanol extract of leaves of dragon scales ( Drymoglossum piloselloides ) effect in reducing the number of Leydig cells , Sertoli cells, germ cells and testis weight, whereas no effect around the seminiferous tubules. This study showed that the ethanol extract of leaves of dragon scales ( Drymoglossum piloselloides ) affect testis weight with the best dose of P4 ( 12.6 mg/200g BB ) and the number of Leydig cells, Sertoli cells and germ cells effectively decreased at doses of P1 ( 5.04 mg / kg ). Yet the circumference of the seminiferous tubules did not decrease significantly.
