Abstract
ABSTRAK
Daun sisik naga (Drymoglossum piloselloides) merupakan salah satu tumbuhan dari golongan Polypodiaceae yang banyak tumbuh di Indonesia. Daun D. piloselloides mengandung senyawa tanin, saponin, steroid dan flavonoid. Senyawa flavonoid dilaporkan dapat bertindak sebagai bahan antifertilitas yang mampu menghalangi aktivitas enzim StAR dan P450ssc dalam mensintesis testosteron di dalam sel leydig. Akibat dari penghambatan tersebut, maka peran testosteron dalam menstimulasi translasi protein testikular akan ikut terganggu. Oleh karena itu, penelitian ini bertujuan untuk mengetahui kadar testosteron dan profil protein testikular tikus putih (Rattus norvegicus) jantan setelah pemberian ekstrak etanol daun sisik naga (D. piloselloides).
Sampel yang digunakan dalam penelitian ini adalah 30 ekor tikus jantan, strain Wistar berumur 2-3 bulan yang dibagi menjadi 5 kelompok perlakuan. Tikus perlakuan 1 (P1) diberi ekstrak etanol daun D. piloselloides sebesar 5,04mg/200g BB tikus, P2:7,56mg/200g BB tikus, P3:10,08mg/200g BB tikus dan P4:12,6mg/200g BB tikus serta perlakuan kontrol (K):0mg/200g BB tikus. Ekstrak etanol daun D. piloselloides diberikan secara oral selama 48 hari untuk kemudian diukur kadar testosteron dan profil protein testikular yang dimiliki tikus putih jantan.
Hasil penelitian menunjukkan bahwa semakin tinggi pemberian ekstrak etanol daun D. piloselloides maka semakin rendah kadar testosteron tikus putih jantan yakni K:1,09ng/ml, P1:1,07ng/ml, P2:0,98ng/ml, P3:0,22ng/ml dan P4:0,2ng/ml, dimana pemberian ekstrak etanol daun D. piloselloides yang efektif dalam menurunkan kadar testosteron adalah P3(10,08mg/200g BB tikus). Sedangkan profil protein testikular setelah pemberian ekstrak etanol daun D. piloselloides adalah 20 pita protein terekpresi pada kontrol, 16 pita protein pada P1, 13 pita protein pada P2 dan 15 pita protein pada P3 dan P4.
ABSTRACT
Sisik naga’s (Drymoglossum piloselloides) leaf is one of a class of Polypodiaceae plants that grow in Indonesia. Leaves of D. piloselloides contains tannin, saponin, steroids and flavonoids. Flavonoids have been reported to act as a material capable of blocking the activity antifertility enzymes StAR and P450ssc in synthesizing testosterone in Leydig cells. As a result of the inhibition, the role of testosterone in stimulating protein translation will be disrupted testicular. Therefore, this study aimed to determine the levels of testosterone and testicular protein profiles of white rats (Rattus norvegicus) males after giving ethanol extract of sisik naga leaves (D. piloselloides).
The sample used in this study were 30 male rats , strain Wistar aged 2-3 months and divided into 5 treatment groups. First treated rat (P1) were given ethanol extract of leaves of D. piloselloides 5,04mg/200g BB rat, P2:7,56mg/200g BB rat, P3:10,08mg/200g BB rats and P4:12,6mg/200g BB rat and the control treatment (K) BB:0mg/200g rat . Ethanol extract of leaves of D. piloselloides administered orally for 48 days and then measured levels of testosterone and testicular protein profiles owned by white male rats .
The results showed that the higher ethanol extract of D. piloselloides leaves give the lower testosterone levels of the male white rat K:1,09 ng/ml, P1:1,07 ng/ml, P2:0,98 ng/ml, P3:0,22 ng/ml and P4:0,2 ng/ml, where the ethanol extract of D. piloselloides leaves effective in lowering testosterone levels was P3 (10,08 mg/200g BB rats). While testicular protein profiles after giving ethanol extract of leaves D. piloselloides is 20 protein bands showed in control, 16 protein bands in P1, 13 protein bands in P2 and 15 protein bands on P3 and P4.
