Abstract
INDONESIA:
Tanaman Alfalfa (Medicago sativa L.) merupakan jenis tanaman legum yang dimanfaatkan sebagai bahan dasar pembuatan produk kesehatan, karena mengandung klorofil empat kali lebih besar dibandingkan tanaman sayur lainnya. Kandungan klorofil dapat diperoleh melalui teknik kultur jaringan salah satunya kultur kalus. Kultur kalus dapat menghasilkan senyawa metabolit sekunder dan dapat digunakan perbanyakan vegetatif. Salah satu faktor penentu keberhasilan kultur kalus tersebut adalah zat pengatur tumbuh yang digunakan. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh perlakuan beberapa konsentrasi 2,4-D yang dikombinasi dengan air kelapa terhadap pertumbuhan dan kandungan klorofil total kalus Alfalfa (Medicago sativa L.) pada media MS.
Penelitian dilakukan di Laboratorium Kultur Jaringan Tumbuhan, Jurusan Biologi, FSAINTEK UIN Malang pada bulan Maret-Mei 2014. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) pola faktorial atau 12 perlakuan. Faktor pertama adalah 2,4-D (0 mg/l; 1 mg/l; 2 mg/l; 3 mg/l), sedangkan faktor kedua adalah air kelapa (10%; 15%; 20%) dengan 4 ulangan. Data yang diperoleh dianalisis dengan mengunakan Analysis of Varian (ANOVA) yang dilanjutkan dengan uji Duncan Multiple Range Test (DMRT) pada taraf uji 95%.
Hasil penelitian menunjukkan bahwa waktu inisiasi kalus tercepat (5,4 hari) diperoleh pada perlakuan 10% air kelapa dengan 3 mg/l 2,4-D. Berat basah kalus tertinggi (0,348850 gram ) diperoleh pada perlakuan 10% air kelapa dengan 2 mg/l 2,4-D. Kandungan klorofil total kalus tertinggi (3,16 x 104 mg/kg) diperoleh pada perlakuan 20% air kelapa dengan 3 mg/l 2,4-D. Persentase tumbuh kalus sebesar 100%, tekstur kalus remah dan warna kalus kuning diperoleh pada semua perlakuan dengan penambahan 2,4-D.

Alfalfa plant (Medicago sativa l.) is a species of legume plants are utilized as the raw material manufacture health products, because they contain chlorophyll is four times greater than other vegetable crops. Duplication of these plants are still experiencing barriers among them, because it doesn’t contain the embryo. These problems can be corrected by using tissue culture techniques, one of them is kalus culture. Kalus culture can produce secondary metabolites compounds and it can be used as vegetative reproduction. One of the success determinants of kalus culture is a growing substance that used. The purpose of this research is knowing the influence of several treatment on several concentrations of 2,4-D were combined with coconut water againts the growth and content of Alfalfa kalus complete chlorophyll (Medicago sativa I.) on MS medium.
Research is conducted in a Laboratory of Plants Tissue Culture, Biology Department, Science and Technology Faculty, Maliki State Islamic University of Malang in March-May 2014. This study used a Rancangan Acak Lengkap (RAL) in factorial pattern or 12 treatment. The first factor is 2,4-D (0 mg/l; 1 mg/l; 2 mg/l; 3 mg/l), whereas the second factor is coconut water (10%; 15%; 20%) with 4 replicates. The obtained data were analyzed using Analysis of Variance (ANOVA) that followed by Duncan Multiple Range Test (DMRT) on the test level of 95%.
The results of research showed that the fastest initiation time of kalus (5.4 days) is obtained at treatment 10% coconut water with 3 mg/l 2,4-D. The highest wet Weight of kalus (0,348850 grams) is obtained at treatment 10% coconut water with 2 mg/l 2,4-D. The highest content of total chlorophyll of kalus (3.16 x 104 mg/kg) is obtained at the treatment 20% coconut water with 3 mg/l 2,4-D. Percentages of kalus growing are 100% texture kalus is crumb and the colour is yellow obtained at all treatment with the addition of 2,4-D.
