Abstract
INDONESIA:
Hemostasis merupakan suatu keadaan kompleks yang mengontrol perdarahan dengan tujuan mempertahankan keenceran darah agar tetap mengalir. Pemicu terjadinya koagulasi yang menyebabkan adanya peningkatan thrombus. (thrombosis) dan sumbatan dapat disebabkan oleh faktor internal maupun eksternal. Efek samping dan permasalahan pengobatan antikoagulan menjadi penyebab pentingnya penemuan alternatif pengobatan antikoagulan dari bahan alam yang dapat memperpanjang waktu pembekuan darah secara aman. Penelitian ini bertujuan untuk memprediksi aktivitas senyawa antikoagulan pada ekstrak etanol 96% herba serai (Cymbopogon citratus) terhadap reseptor (6WV3) dan (2P16). Senyawa hasil metabolite profiling metode UPLQ-QToF MS/MS ekstrak etanol 96% herba serai (Cymbopogon citratus) dianalisis sifat fisikokimia dengan SwissADME dan toksisitas menggunakan ProTox II online tools. Senyawa terpilih dengan sifat fisikokimia dan toksisitas yang baik dipreparasi menggunakan ChemDraw Ultra 12.0.2, lalu dioptimasi geometri dengan Avogadro 1.2.0, kemudian dilakukan molecular docking dengan PyRx 0.8 metode Autodock Vina. Visualisasi hasil molecular docking dilakukan dengan Biovia Discovery Studio 2021 untuk mencari interaksi antagonis terhadap warfarin maupun apixaban. Penambatan dikatakan validasi software tersebut dapat digunakan karena nilai RMSD <2A Didapatkan 19 hasil senyawa metabolite profiling memenuhi aturan sifat fisikokimia baik dan tidak toksik. Analisis secara in silico terhadap protein 6WV3 dan tidak toksik. Analisis secara in silico terhadap protein6WV3 dan 2P16 menunjukan terdapat satu senyawa yang memberikan interaksi antagonis dengan kemiripan jarak farmakofor dan ikatan asam amino native ligan warfarin. Senyawa -Deoxy-2-({[(2-methyl-2- propanyl)oxy]carbonyl}amino)-D- glucopyranose golongan senyawa alkaloid yang diprediksi memiliki potensi yang paling baik sebagai antikoagulan kemiripan parameter docking dengan native ligan warfarin dan apixaban. Oleh karena itu, senyawa ektrak etanol 96% herba (Cymbopogon citratus) diprediksi memiliki aktivitas antikoagulan dengan mekanisme penghambatan

Hemostasis is a complex condition that controls bleeding with the aim of maintaining the thinness of the blood to keep it flowing. The trigger for hypercoagulation which causes an increase in thrombus (thrombosis) and blockage can be caused by internal or external factors. The side effects and problems of anticoagulant treatment are the cause of the importance of finding alternative anticoagulant treatments from natural ingredients that can safely prolong blood clotting time. This study aims to predict the activity of anticoagulant compounds in the 96% ethanol extract of lemon grass (Cymbopogon citratus) against receptors (6WV3) and (2P16). Compounds resulting from metabolite profiling using the UPLQ-QToF MS/MS ethanol extract 96% lemongrass (Cymbopogon citratus) were analyzed for physicochemical properties using SwissADME and toxicity using ProTox II online tools. Selected compounds with good physicochemical and toxicity properties were prepared using ChemDraw Ultra 12.0.2, then optimized geometry with Avogadro 1.2.0, then molecular docking was carried out with PyRx 0.8 Autodock Vina method. Visualization of molecular docking results was carried out with Biovia Discovery Studio 2021 to look for antagonistic interactions with warfarin and apixaban. The mooring said that the validation software could be used because the RMSD value was <2A. 19 results of the profiling metabolite compounds met the rules for good physicochemical properties and were non-toxic. In silico analysis of 6WV3 protein and non-toxic. In silico analysis of 6WV3 and 2P16 proteins showed that there is one compound that provides antagonistic interactions with similar pharmacophore distances and native amino acid binding of warfarin ligands. The compound 2-Deoxy-2-({[(2-methyl-2- propanyl)oxy]carbonyl}amino)-D-glucopyranose belongs to the alkaloid compound that is predicted to have the best potential as an anticoagulant, with similar docking parameters to the native ligands warfarin and apixaban. Therefore, 96% herbal ethanol extract compound (Cymbopogon citratus) is predicted to have anticoagulant activity with an inhibitory mechanism.
