Abstract
ABSTRAK:
Biofilm merupakan kumpulan bakteri interaktif yang melekat satu sama lain pada substrat biologis maupun non-biologis yang dilindungi dalam matriks eksopolisakarida (EPS). Adapun bakteri yang berada di dalam matriks eksopilisakarida menyebabkan terjadinya perlindungan bakteri dari mekanisme imun pejamu. Klebsiella pneumoniae merupakan bakteri yang mampu membentuk biofilm. Pembentukan biofilm melalui 5 tahap, yaitu perlekatan sel, adhesi sel ke sel yang lain, proliferasi dan pertumbuhan sel, pematangan, dan pelepasan atau penyebaran. Aspergillus terreus mengandung metabolit sekunder, seperti terreic acid dan mengandung enzim seperti, enzim selulase dan protease sehingga diharapkan CFS A. terreus mampu mengganggu pertumbuhan biofilm. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibiofilm A. tereus terhadap biofilm K.pneumoniae. Penelitian ini menggunakan Metode Tissue Culture Plate (TCP) pada microplate dengan 7 kelompok perlakuan dengan kategori dua kelompok control, yaitu positif dan negative, serta lima kelompok uji dengan masing-masing 4 kali pengulangan. Adapun onsentrasi CFS A. terrreus yang diujikan, yaitu 100%, 50%, 25%, 12,5%, dan 6,25%. Uji antibiofilm yang digunakan meliputi uji pencegahan perlekatan biofilm, uji penghambatan pertumbuhan biofilm, dan uji kerusakan/penghancuran biofilm. Persentase aktivitas antibiofilm didapatkan melalui rumus yang telah dimasukkan nilai Optical Density (OD) tiap uji. Aktivitas antibiofilm A. terreus terhadap pencegahan perlekatan, penghambatan pembentukan, dan kerusakan/penghancuran biofilm K. pneumoniae didapatkan nilai tertinggi pada konsentrasi CFS A. terreus 100%. Berdasarkan hasil tersebut, disimpulkan bahwa pemberian konsentrasi tersebut, CFS A. terreus memiliki aktivitas antibiofilm terhadap biofilm K. pneumoniae.
ABSTRACT:
Biofilm is a collection of interactive bacteria that attached to each other on biological and non-biological substrates and protected in an exopolysaccharide matrix (EPS). The bacteria that are in the exopilisaccharide matrix cause bacterial protection from the host immune mechanism. Klebsiella pneumoniae is a bacterium capable of forming biofilm. There are 5 stages of forming a biofilm, namely cell attachment, cell adhesion to other cells, cell proliferation and growth, maturation, and through dispersal. Aspergillus terreus contains secondary metabolites, such as terreic acid and enzymes such as cellulase and protease enzymes, so it is hoped that CFS A. terreus will be able to interfere with biofilm growth. The purpose of this study was to determine the antibiofilm activity of A. tereus against biofilm of K. pneumoniae. This study used the Tissue Culture Plate (TCP) method on microplate with 7 treatments, there are two control groups, namely positive and negative, and five test groups with 4 different categories. The concentrations of CFS A. terrreus tested were 100%, 50%, 25%, 12.5%, and 6.25%. Antibiofilm tests used include biofilm adhesion prevention tests, biofilm growth tests, and biofilm damage/destruction tests. The percentage of antibiofilm activity was obtained through a formula that included the Optical Density (OD) value for each test. The highest value antibiofilm activity of A. terreus on prevention of attachment, inhibition of formation, and damage/destruction of K. pneumoniae biofilm was obtained at 100% A. terreus CFS concentration. The results is that present CFS A. terreus has antibiofilm activity against K. pneumoniae biofilm.
