Abstract
ABSTRAK:
Escherichia coli (E. coli) merupakan bakteri gram negatif yang menyebabkan infeksi saluran kemih (ISK) dan memiliki kemampuan dalam membentuk biofilm. Terbentuknya biofilm menyebabkan terapi ISK menjadi kurang efektif. Aspergillus oryzae mengandung enzim amilase dan enzim protease yang dapat digunakan sebagai antibakteri dan antibiofilm. Penelitian ini bertujuan untuk menguji aktivitas antibakteri dan antibiofilm Aspergillus Oryzae terhadap Escherichia coli. Uji aktivitas antibiofilm menggunakan uji penghambatan pertumbuhan biofilm/Minimum Biofilm Inhibitory Concentration (MBIC). Uji Kosentrasi Hambat Minimum. (KHM) dan Kosentrasi Bunuh Minimum (KBM) bertujuan untuk mengetahui kemampuan Aspergillus oryzae dalam menghambat dan membunuh pertumbuhan bakteri Escherichia coli. Metode penelitian menggunakan metode Microtiter Plate Biofilm Assay. Hasil uji aktivitas antibiofilm menunjukkan bahwa Aspergillus oryzae memiliki aktivitas penghambatan pertumbuhan biofilm dengan persentase aktivitas terbesar secara berturut-turut pada 100%, 50%, 25%, 12,5% dan 6,25%. Hasil uji KHM dan KBM menunjukkan Aspergillus oryzae tidak mampu menghambat dan membunuh bakteri Escherichia coli.
ABSTRACT:
Escherichia coli (E. coli) is a gram-negative bacterium that causes urinary tract infections (UTIs) and has the ability to form biofilms. The formation of biofilms makes UTI’s therapy less effective. Aspergillus oryzae contains amylase enzymes and protease enzymes that can be used as antibacterials and antibiofilms. The study aimed to test the antibacterial and antibiofilm activity of Aspergillus Oryzae against Escherichia coli. Antibiofilm activity tests use Minimum Biofilm Inhibitory Concentration (MBIC). Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) tests aim to determine the ability of Aspergillus aculeatus to inhibit and kill the growth of Escherichia coli bacteria. The research method uses the Microtiter Plate Biofilm Assay method. The results of antibiofilm activity tests showed that Aspergillus oryzae had the inhibition activity of biofilm growth with the largest percentage of activity in a row at 100%, 50%, 25%, 12.5% and 6.25%. MIC and MBC test results showed Aspergillus oryzae was unable to inhibit and kill Escherichia coli bacteria.
