Kombinasi ekstrak daun pegagan (Centella asiatica (L) urban) dan beluntas (Plucea indica.(L)Urban) terhadap jumlah folikel, kadar SOD dan MDA ovarium tikus putih (Rattus norvegicus)

Pegagan (Centella asiatica (L) Urban dan beluntas (Plucea indica (L) Urban merupakan bahan alam yang digunakan sebagai obat tradisional yang mengandung bahan aktif golongan triterpen, alkaloid, tanin dan flavonoid yang mampu mempengaruhi organ reproduksi betina. Pegagan dan beluntas diduga mempengaruhi kerja hormon reproduksi, sehingga mempengaruhi pematangan folikel ovarium. Kedua bahan tersebut juga memiliki kadar antioksidan yang tinggi. Diharapkan kedua bahan tersebut menghasilkan dosis dengan efek yang menguntungkan bagi sistem reproduksi.

Penelitian ini merupakan penelitian eksperimental yang menggunakan rancanagn acak lengkap (RAL) dengan 6 perlakuan dan 4 ulangan.Perlakuan digunakan ekstrak daun pegagan dan beluntas dengan P0 (kontrol), P1 (dosis 25 mg/Kg BB), P2 (dosis 50 mg/Kg BB),P3 (dosis 75 mg/Kg BB),P4 (dosis 125 mg/Kg BB), dan P5 (dosis 200 mg/Kg BB). Hewan yang digunakan adalah tikus betina fertil galur wistar berumur 2 bulan, dengan lama perlakuan 15 hari. Parameter dalam penelitian ini meliputi jumlah folikel primer, sekunder, tersier, de Graff, dan korpus luteum serta kadar SOD san MDA ovarium. Data dianalisis dengan Anova dan jika terdapat pengaruh maka dilanjut dengan uji BNT 5%.

Berdasarkan hasil analisis statistikmenunjukkan ekstrak daun pegagan (Centella asiatica (L) Urban dan beluntas (Plucea indica (L) Urban berpengaruh terhadap jumlah folikel ovarium primer, dan tidak ada pengaruh nyata terhadap folikel sekunder, tersier, de graff dan korpus luteum. Sedangkan pegagan dan beluntas menunjukkan adanya pengaruh nyata pada kadar SOD dan MDA. Dosis yang menunjukkan kadar SOD ovarium tertinggi adalah dosis 200 mg/Kg BB, sedangkan MDA yang paling rendah adalah dosis 200 mg/Kg BB.

Pegagan (Centellaasiatica (L) Urban and beluntas(Plucea indica (L) Urban are natural matters used as traditional medicine containing active substance, such as triterpen, alkaloids, tannins and flavonoids that usable to affect female reproductive organs. Pegagan and beluntas expected to affect reproductive hormones working, so it can affect the maturation of ovarian follicles. They also have high antioxidant levels. So, both of themexpected to produce a dose with a beneficial effect for reproductive system.

This research is experimental research using random complete program (RCP) with 6 treatments and four replication. Treatment used pegagan leaf extract and beluntas with P0 (control), P1 (25 mg/KgBB dose), P2 (50 mg/KgBB dose), P3 (75 mg/KgBB dose), P4 (125 mg/KgBB dose) and P5 (200 mg/KgBB dose). Animal trial that used was female wistar strain rats fertile 2 months old, with 15 days treatment. The parameters in this study includes the total of the primary follicle, secondary, tertiary, de Graff, and the corpus luteum and also contents of SOD san MDA ovaries. Data was analyzed by Anova and if influence was found, it will be continued with BNT 5% test.

Based on the statistical analysis results that showedpegagan leaf extract (Centella asiatica (L) Urban and beluntasleaf extract (Plucea indica (L) Urban influenceto primary ovarian follicle total, and didn’t influence reallyto secondary, tertiary follicles, de graff and the corpus luteum. Whereas pegagan and beluntas showed a real influence on the SOD and MDAcontents. Dose showed the highest SOD ovarian contents was 200 mg/Kg BB dose, while MDA was the lowest one with 200 mg/KgBB dose.
