Abstract

INDONESIA:

Katuk (Sauropus androgynus (L.) Merr) merupakan salah satu tanaman herbal yang mempunyai banyak manfaat seperti untuk pelancar ASI, obat demam, obat bisul, dan darah kotor. Pemanfaatan daun katuk sebagai obat tradisional dalam jangka waktu yang panjang, memungkinkan adanya efek toksik. Toksikan biasanya terakumulasi di hepar sebagai organ yang berfungsi untuk sebagai mendetoksifikasi toksikan yang masuk ke dalam tubuh. Gangguan fungsi hepar dapat dilihat dengan pengujian biokimia terhadap kadar enzim transaminase AST dan ALT yang diproduksi oleh hepatosit. Tujuan penelitian adalah untuk mengetahui hasil uji toksisitas subkronik ekstrak air daun katuk (Sauropus androgynus (L.) Merr) terhadap kadar enzim transaminase (AST dan ALT) hepar tikus (Rattus norvegicus) betina serta mengetahui kadar enzim transaminase (AST dan ALT) pada hepar tikus (Rattus norvegicus) betina yang diberi perlakuan tersebut.

Penelitian ini merupakan penelitian eksperimental laboratorium yang menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 6 ulangan yang diujikan pada 24 ekor tikus. Beberapa perlakuan tersebut adalah tikus diberi ekstrak air daun katuk dengan dosis 0mg/KgBB (P0), dosis 45 mg/KgBB (P1), dosis 60 mg/KgBB (P2), dan dosis 75 mg/KgBB (P3). Hewan coba yang digunakan adalah tikus betina galur wistar yang berumur 2 bulan. Parameter yang diamati adalah kadar enzim transaminase (AST dan ALT) yang terdapat pada organ hepar. Data hasil penelitian dianalisis menggunakanan statistik uji normalitas, uji homogenitas dan uji One Way ANOVA dengan F tabel 5%.

Hasil penelitian menunjukkan berdasarkan uji ANOVA nilai p ALT= 0.135 dan nilai p AST= 0.393 karena nilai p > 0.05. maka hasil uji tersebut bermakna bahwa ekstrak air daun katuk (Sauropus androgynus (L.) Merr) tidak berpengaruh toksik terhadap kadar enzim transaminase (ALT dan AST) hepar tikus (Rattus norvegicus) betina. Aktivitas antioksidan ekstrak air daun katuk secara statistik tidak mempengaruhi kadar enzim transaminase (ALT dan AST).

ENGLISH:

Katuk (Sauropus androgynus (L.) Merr) is a herbal plant that has much benefits such as to induce the secretion of Mother’s Breast Milk (ASI), fever drug, ulcers, medication and blood dirty drug. Katuk leaves utilisation as a traditional medicine in a long period of time, allowing the presence of toxic effects. Toxican usually accumulates in hepar as an organ that serves to detoxify as tokxican that goes into the body. Malfunctioning of hepar can be seen with biochemical testing against the levels of transaminase enzyme (ALT and AST) are produced by hepatocytes. The purpose of the research is to find out the results of subcronic toxicity test of leaves katuk (Sauropus androgynus (L.) Merr) aqueous extract towards the levels of transaminase enzyme (ALT and AST) hepar rat (Rattus norvegicus) females as well as find out the levels of transaminase enzymes (ALT and AST) in hepar rat (Rattus norvegicus) females who were given the treatment.

This research is experimental research laboratory that uses a completely Randomized Design (CRD) with 4 treatments and 6 replication which is examined at 24 rats. Some of the rat given the treatment Katuk leaves (Sauropus androgynus (L.) Merr) aqueous extract followed by administration a dose of 0 mg/KgBB (P0), a dose of 45 mg/KgBB (P1), a dose of 60 mg/KgBB (P2), and a dose of 75 mg/KgBB (P3). The animals are trying to use are strains of wistar female rats aged 2 months. The parameters observed were the levels of transaminase enzyme (ALT and AST) in organ hepar. Data research results are analyzed using statistical test of normality, homogeneity test and One Way ANOVA test with F table 5%.

The results showed based on ANOVA test value p ALT = 0.135 and the value of the p AST = 0.393 because the value (p > 0.05). then the test result means that the aquous extract of leaves of (Sauropus androgynus (L.) Merr) has no effect against the toxic levels of transaminase enzymes (ALT and AST) hepar rat (Rattus norvegicus) females. Antioxidant activity katuk leaves (Sauropus androgynus (L.) Merr) aqueous extracts of statistically does not affect levels of transaminase enzymes (ALT and AST).
