Pengaruh pemberian ekstrak rimpang temu ireng (curcuma aeruginosa roxb) terhadap skor ulkus peptikum yang diamati secara makroskopis pada tikus putih (rattus novergicus) strain wistar yang diinduksi indometasin

ABSTRAK:

Latar Belakang: Ulkus peptikum merupakan suatu penyakit pada lambung dan bagian duodenum proksimal yang ditandai dengan rusaknya lapisan mukosa, submukosa hingga lapisan otot akibat ketidakseimbangan antara fungsi proteksi dengan faktor perusak. Penyebab tersering terjadinya ulkus peptikum diantaranya infeksi H.pylory dan penggunaan NSAIDs. Salah satu tanaman yang berpotensi sebagai gastroprotektif dan antioksidan dalam menurunkan gejala ulkus peptikum ialah rimpang temu ireng (Curcuma aeruginosa Roxb). Tujuan penelitian ini adalah menguji efek gastroprotektif dan antioksidan ekstrak rimpang temu ireng pada lambung tikus yang diinduksi Indometasin. Metode: Penelitian ini menggunakan 30 ekor tikus, dibagi kedalam 6 kelompok. Kelompok I adalah kelompok normal. Kelompok II kontrol negatif (Induksi indometasin 30mg/kgBB). Kelompok III kontrol positif (pemberian omeprazole 20mg/kgBB). Kelompok IV, V dan VI diberi ekstrak rimpang temu ireng, dengan dosis bertingkat yaitu 150, 300 dan 450 mg/kgBB. Perlakuan dilakukan selama 14 hari, pada hari ke-14 tikus diinduksi Indometasin lalu dibedah. Data penelitian skor ulkus peptikum dianalisis dengan uji Saphiro-Wilk dan uji Levene, selanjutnya di uji menggunakan Kruskall Wallis dengan derajat kepercayaan 95%. Hasil uji statistik perlakuan dosis 300 mg/kgBB dengan kontrol negatif menunjukkan adanya perbedaan signifikan dengan nilai p 0,005. Berdasarkan hasil penelitian dapat disimpulkan bahwa pemberian ekstrak rimpang temu ireng dapat menurunkan skor ulkus peptikum yang diamati secara makroskopis pada tikus putih strain wistar yang diinduksi Indometasin.

Background: Peptic ulcer is a disease of the stomach and the proximal duodenum which is characterized by damage to the mucosal layer, submucosa to the muscle layer due to the balance between protective functions and damaging factors. The most common causes of peptic ulcers include H. pylori infection and the use of NSAIDs. One of the plants that have gastroprotective and antioxidant effect in reducing peptic ulcer symptoms is temu ireng (Curcuma aeruginosa Roxb) rhizome. The purpose of this study was to examine the gastroprotective and antioxidant effects of temu ireng rhizome extract on the stomach of rats induced by Indomethacin. Methods: This study used 30 rats, divided into 6 groups. Group I is the normal group. Group II negative control (Induced with indomethacin 30mg/kgBW). Group III was positive control (with omeprazole 20mg/kgBW). Groups IV, V and VI were given extracts of temu ireng rhizome, with graded doses of 150, 300 and 450 mg/kgBW. The treatment was carried out for 14 days, on the 14th day the rats were induced by Indomethacin and then dissected. Research data on peptic ulcer scores were analyzed using the Saphiro-Wilk test and Levene's test, then tested using Kruskall Wallis with a 95% confidence degree. The results of the statistical test for treatment of 300 mg/kgBW with a negative control showed a significant difference with a p value of 0.005. Based on the results of the study, it can be concluded that the administration of rimpang temu ireng extract can reduce the score of peptic ulcers observed macroscopically in indomethacin-induced
