Abstract
ABSTRAK:
Oral Squamous Cell Carninoma (OSCC) merupakan tumor ganas, yang mampu berinfiltrasi, berasal dari jaringan epitel, dan dapat dicetuskan oleh karsinogen yang dapat menyebabkan adanya mutasi somatik DNA sehingga akan meningkatkan kadar sitokin pro-inflamasi salah satunya TNF-α, Tumor Necrosis Factor-Alpha (TNF-α) dapat meningkatkan progresifitas dan perburukan pada OSCC. Daun krisan (Chrysantemum cinerariifolium) mengandung senyawa terpenoid dan flavonoid yang memiliki aktivitas anti-inflamasi, modulasi enzim ROS, induksi apoptosis, autophagy, menekan proliferasi dan invasi sel kanker, sehingga diharapkan ekstrak daun krisan (Chrysantemum cinerariifolium) dapat menurunkan kadar TNF-α untuk mencegah progresifitas dan perburukan OSCC. Tujuan penelitian ini adalah untuk mengetahui ekstrak daun krisan dalam menurunkan kadar TNF-α pada OSCC. Penelitian ini dilakukan pada tikus Sprague Dawley jantan yang diinduksi DMBA 0,5% dan dibagi menjadi 5 kelompok, yaitu kelompok kontrol positif yang diinduksi DMBA tanpa diberi terapi ekstrak, kontrol negatif tanpa induksi DMBA dan tanpa pemberian ekstrak daun krisan, lalu kelompok perlakuan yang diinduksi dengan DMBA lalu diberi terapi ekstrak daun krisan yang dibagi menjadi 3 yaitu kelompok dosis 1 (50 mg/KgBB), dosis 2 (100 mg/KgBB), dosis 3 (200 mg/KgBB). Ekspresi TNF-α didapatkan dengan melakukan pengecatan imunohistokimia, selanjutnya TNF-α yang terekspresi diamati menggunakan mikroskop dan dihitung jumlahnya pada masing-masing kelompok. Hasil pengamatan dan penelitian menunjukkan bahwa kadar TNF-α a pada kelompok kontrol positif yaitu dengan rata-rata 56,75, pada kelompok kontrol negatif yaitu 9,65, kelompok perlakuan dosis 1 yaitu 45,125, kelompok perlakuan dosis 2 25,025 dan kelompok dosis 3 yaitu 16,4, kemudian kadar TNF-α diuji menggunakan One Way ANOVA yang menunjukkan hasil signifikan (p<0,05), dengan uji Post Hoc Tukey menunjukkan hasil berbeda signifikan antar kelompok. Berdasarkan hasil tersebut disimpulkan bahwa pemberian ekstrak daun krisan dapat menurunkan kadar TNF-α pada OSCC.
Oral Squamous Cell Carninoma (OSCC) is a malignant tumor, capable to infiltrating to
another organs, came from epithelial tissue, which can be triggered by carcinogens that can cause somatic DNA mutations that will increase levels of pro-inflammatory cytokines,including TNF-α, TNF-α may increase progression and worsening of OSCC.
Chrysanthemum leaves (Chrysantemum cinerariifolium) contain Terpenoid and Flavonoid
compounds which have anti-inflammatory activity, modulation of ROS enzymes, induction
of apoptosis, autophagy, suppressing proliferation and invasion of cancer cells, so it’shoped that chrysanthemum leaf extract (Chrysantemum cinerariifolium) can reduceTNFα levelfor prevent progression and worsening of OSCC.
The purpose of this studywas to determine the effectof chrysanthemum leafextract to decrease TNF-αexpression inOSCC.
This research wasconducted onmale SpragueDawley rats induced by DMBA 0,5% which
were divided into 5 groups, there is positive control group induced by DMBA without
chrysanthemum leaf extract therapy, negative control group without DMBA induction and
without chrysanthemum leaf extract therapy, Then treatment group that was induced by
DMBA and given chrysanthemum leaf extract for therapy which was divided into 3 group,
there is dose group 1 (50 mg / KgBB), dose group 2 (100 mg / KgBB), dose group 3 (200
mg / KgBB). TNF-α expression was obtained by imunohistochemical staining, then the
TNF-α expression was observed using a microscope and the amount in each group was
counted by cuonter. The results on observations and research showed that the levels of
TNF-a in positive control group average is 56,75, and negative control group was 9,65,
then the treatment group dose 1 was 45,125, the treatment group dose 2 was 25,025 and the
dose group 3 was 16,4 , then TNF-α levels were tested using One Way ANOVA which
showed significant results (p <0.,5), and significantly different between groups with Post
Hoc Tukey’s test. Based on these results, it was concluded that Chrysanthemum leaf extract
can decrease TNF-α levels in OSCC.
