Abstract
INDONESIA:
Daun Widuri (Calotropis gigantea) merupakan salah satu tanaman yang banyak dimanfaatkan masyarakat sebagai obat tradisional, seperti untuk obat sakit gigi, penawar racun, obat masuk angin dan batuk asma. Namun tanaman ini juga dapat dimanfaatkan sebagai obat antikanker. Daun widuri mengandung bahan aktif kardenolida yang memiliki sifat sitotoksik terhadap sel kanker termasuk kanker fibrosarkoma. Bahan aktif tersebut mampu mengendalikan mitokondria (organ sel penghasil energi) yang dibutuhkan sel kanker untuk berkembang. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol daun widuri (Calotropis gigantea) terhadap gambaran histologis fibrosarkoma mencit (Mus musculus) jantan yang diinduksi 7,12-dimetilbenz(a)antrasena (DMBA) secara in vivo.
Penelitian ini bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan 6 perlakuan 5 kali ulangan. Perlakuan yang digunakan adalah (K-) Kontrol Negatif, (K+) Kontrol Positif, (P1) dosis 50 mg/kg BB, (P2) dosis 100 mg/kg BB, (P3) dosis 150 mg/kg BB, (P4) Metotrexat dosis 2,5 mg/kg BB serta 7,12-dimetilbenz(a)antrasena (DMBA) 25 mg/Kg BB. Hewan yang digunakan mencit jantan sebanyak 30 ekor yang berumur ±50-60 hari. Data hasil penelitian meliputi jumlah sel fibroblas dan persentase luas kerusakan serat kolagen mencit (Mus musculus). Data yang diperoleh dianalisis dengan ANOVA satu arah, apabila terdapat perbedaan sangat nyata maka dilanjutkan dengan uji duncan.
Hasil penelitian menunjukkan bahwa ekstrak daun widuri (Calotropis gigantea) berpengaruh terhadap histologi fibrosarkoma mencit (Mus musculus), yang memperlihatkan bahwa ekstrak daun widuri (Calotropis gigantea) mampu menurunkan jumlah sel fibroblas dan memperbaiki serat kolagen yang mengalami kerusakan. Ekstrak daun widuri pada dosis 150 mg/kg BB lebih baik daripada obat kanker sintesis yaitu Metotrexat dosis 2,5 mg/kg BB.
ENGLISH:
Calotropis gigantean leaf is one of many plants which are used by people as traditional medicine, for example as toothache medicine, poison antidote, feverish medicine and asthma cough. However this plant also can used as cancer medicine. Calotropis gigantea contains compound cardenolide which have cytotoxic concerning cancer cell include fibrosarcoma. That actif substance can restrain mitocondria (cell organ energy producer) which is needed by cancer cell to develop. This research as a purpose to know influence ethanol extract of calotropis gigantea concerning histology image of male mice (mus musculus) which is induced 7,12-dimetilbenz(a)antrasena (dmba) in vivo.
This research is experimental which use Complete Random Plan (CRP) with 6 treatment 5 times repetition. The treatment which used is (K-) Negative Control, (K+) Positive Control, (P1) dosage 50 mg/kg BB, (P2) dosage 100 mg/kg BB, (P3) dosage 150 mg/kg BB, (P4) Metotrexat dosage 2,5 mg/kg BB and 7,12-dimetilbenz(a)antrasena (DMBA) 25 mg/Kg BB. The animal which used is 30 male mice attain the age of ±50-60 days. Data result od research comprise sum of fibroblast cell and wide ravage percentage of male mice (Mus musculus) colagent fiber. Data which found then analysis with ANOVA one way, when be found very real diverification then continued with Duncan experiment.
The result of reseacrh indicate that calotropis gigantea extract influential concerning fibrosarcoma histology of male mice (Mus musculus), which show that calotropis gigantean extract capable to reduce sum of fibroblast cell and repair colagent fiber which broken. Calotropis gigantea extract at dosage 150 mg/kg BB better than synthesis cancer medicine that is Metotrexat dosage 2,5 mg/kg BB.
