Abstract

INDONESIA:

Penyakitikan yang disebabkan bakteri Aeromonashydrophilla adalah septisemia. Penyakit streptococcosis disebabkan oleh Streptococcus agalactiae. Kedua bakteri pathogen virulensinyat inggisehingga membutuhkan anti bakteri yang dapat menghambatnya. Temulawak memiliki senyawa kurkumin, alkaloid, flavonoid, saponin, triterpenoid. Senyawa tersebut dapat dimanfaatkan sebagai anti bakterijadi, diduga bakteri endofitnya juga memiliki senyawa tersebut. Terutama kurkumin yang hanya terdapat pada tanaman Curcuma. Perlu diteliti ada tidaknya kurkumin dengan melihat enzim penghasil kurkumin dengan bioinformatika, teknik BLAST.

Penelitian ini merupakan penelitian eksperi mendaneksplorasi. Mengujii solat bakteri endofitrimpang temulawak terhadap bakteri AeromonashydrophilladanStreptococcus agalactiae dengan metodedifusikertassecara in vitro. Analisis data dengan mengukur zona hambatnya. Mengkaji isolat bakteri endofit rimpang temulawak sebagai penghasil enzim kurkupmin dengan bioinformatika. Analisis data dengan melihatpersentasequery cover ≥ 80% dan identity ≥ 30%.

Hasil uji anti bakteri, bakteri endofitrimpang temulawak dapat menghambat bakteri pathogen penyebab penyakit ikan. Zona hambat terbesar pada isolat Pseudomonas stutzeri, 5,5 mm terhadap bakteri Streptococcus agalactiae dan 5,6 mm terhadap Aeromonashydrophilla. Hasil BLAST menunjukkansekuen bakteriendofit dengan enzimpenghaislkurkumin tidak homolog sehingga, dinyatakan bakteri endofit tidak memiliki enzoimpeng hasil kurkumin

ENGLISH:

Fish disease caused by the bacteria Aeromonashydrophilla is septicemia. Streptococcosis disease is caused by Streptococcus agalactiae. Both the high virulence of pathogenic bacteria and thus require antibacterial that can hinder. Wild Ginger has a compound curcumin, alkaloids, flavonoids, saponins, triterpenoids. The compounds can be used as antibacterial so, allegedly the endophitic bacteria also have these compounds; especially, curcumin which is only found in Curcuma. So, it needs to be examined whether there is curcumin to see the enzyme producer curcumin with bioinformatics, using BLAST.

This study is experimental research and exploration. Testing of endophytic bacteria isolates ginger rhizome against bacteria Aeromonashydrophilla and Streptococcus agalactiae with paper diffusion method in vitro. Analysis of the data is done by measuring the inhibitor zone. Assessing the endophytic bacteria isolates ginger rhizome as a producer of enzymes curcumin with bioinformatics. Analysis of the data by looking at the percentage of query cover ≥ 80% and ≥ 30% identity.

Antibacterial test results, ginger rhizome endophytic bacteria can inhibit pathogenic bacteria causing fish diseases. The largest inhibition zone is isolate of Pseudomonas stutzeri, 5.5 mm against the bacteria Streptococcus agalactiae and 5.6 mm against Aeromonashydrophilla. BLAST results show that the sequence of endophytic bacteria and enzymes are not the same. Therefore, otherwise endophytic bacteria do not have a curcuminsintaseenzym.
