Abstract
INDONESIA:
Vitamin E (a-tocoferol) merupakan antioksidan yang melindungi Poly Unsaturated Faty Acids (PUFA) dan komponen membran sel lain dari oksidasi radikal bebas. Vitamin ini mampu meredam kelebihan radikal bebas di dalam sel yang disebabkan oleh berbagai bahan kimia yang masuk ke dalam sel. Salah satu bahan kimia tersebut adalah etanol. Penelitian ini bertujuan untuk mengetahui pengaruh vitamin E (α- tocoferol) terhadap kerusakan, viabilitas, dan abnormalitas kultur primer sel paru-paru fetus hamster yang dipapar etanol.
Jenis penelitian yang dilakukan adalah penelitian eksperimental menggunakan Rancangan Acak Lengkap (RAL)dengan 7 perlakuan yaitu kontrol negatif, kontrol positif, vitamin E (α-tocoferol) konsentrasi 25 µM, 50 µM, 75 µM, 100 µM dan 125 µM yang dipapar etanol 10 mM selama 24 jam dan masing-masing diulang 3 kali. Sampel dalam penelitian ini adalah sel paru-paru fetus hamster umur 2 hari yang dikultur dalam media DMEM 20% FBS dan diberi perlakuan vitamin E dengan konsentrasi yang berbeda, kemudian diinkubasi dalam inkubator CO2 5% suhu 37°C selama 5 hari. Selanjutnya sampel dikultur kembali dan dipapar dengan etanol 10 mM selama 24 jam. Pada 24 jam setelah pemaparan dengan etanol, kultur primer sel paru-paru fetus hamster diamati kerusakan, viabilitas, dan abnormalitas sel.
Hasil penelitian menunjukkan bahwa vitamin E (α-tocoferol) berpengaruh terhadap kerusakan, viabilitas dan abnormalitas kultur primer sel paru-paru fetus hamster yang dipapar etanol. Hasil penelitian menunjukkan bahwa konsentrasi vitamin E (α-tocoferol) yang berpengaruh terhadap kerusakan, viabilitas, dan abnormalitas tersebut adalah konsentrasi 125 µM, 100 µM dan 25 µM.
ENGLISH:
Vitamin E (a-tocoferol) is an antioxidant that protects unsaturated Poly Faty Acids (PUFAs) and other cell membrane components from oxidation by free radicals. This vitamin is capable of reducing excess free radicals in the cells caused by various chemicals that enter the cell. One chemical is ethanol. This study aims to determine the effect of vitamin E (α-tocoferol) against damage, viability, and abnormalities of primary cultured fetal hamster lung cells exposed to ethanol.
This type of research is carried out experimental studies using Complete Randomized Design with 7 treatment of the negative control, positive control, vitamin E (α-tocoferol) concentration of 25 μM, 50 μM, 75 μM, 100 μM and 125 μM are exposed to ethanol 10 mM for 24 hours and each was repeated three times. The sample in this study are fetal hamsters lung cell aged 2 days were cultured in DMEM medium 20% FBS and vitamin E were treated with different concentrations, then incubated in 5% CO2 incubator temperature of 37°C for 5 days. Further samples were cultured again and exposed to 10 mM ethanol for 24 hours. At 24 hours after exposure to ethanol, the primary cell cultures of fetal hamster lung damage observed, viability, and cell abnormalities.
The research results show that vitamin E (α-tocoferol) affect the damage, viability and abnormalities of primary cultured fetal hamster lung cells exposed to ethanol. The research results show that concentrations of vitamin E (α-tocoferol) are effective against damage, viability, and abnormality was concentration of 125 μM, 100 μM and 25 μM.
