Abstract
INDONESIA:
Daerah bekas tambang mengandung zat pencemar berupa logam berat Pb yang dapat menyebabkan terganggunya ekosistem dan kesehatan manusia. Salah satu upaya untuk menangani hal tersebut yaitu menggunakan metode bioremediasi dengan memanfaatkan bakteri. Penelitian ini bertujuan untuk mengidentifikasi bakteri berdasarkan morfologi dan melihat ketahanannya terhadap logam Pb dalam berbagai konsentrasi. Tujuh isolat yang dihasilkan dari hasil isolasi bakteri yaitu T2P1a, T2P1b, T2P2, T3P1, T3P2, T3P3, dan T3P4. Bentuk morfologi yang dihasilkan dari seluruh isolat rata-rata berbentuk bulat, berwarna putih kecoklatan, berukuran kecil, memiliki tepi rata dan permukaan yang timbul. Jumlah koloni terbanyak pada Telaga 2 dan Telaga 3 berturut-turut dihasilkan oleh T2P2 dan T3P2 dengan jumlah koloni sebanyak 1,3 x 108 dan 3,3 x 1010 cfu/mL yang ditumbuhkan pada media padat tanpa adanya kandungan logam Pb dengan masa inkubasi 2x24 jam pada suhu ruang. Ketahanan bakteri terhadap berbagai konsentrasi logam berdasarkan uji minimum inhibitory concentration (MIC) menunjukkan bahwa seluruh isolat pada konsentrasi Pb 6 mM rata-rata tidak menghasilkan diameter zona hambat (0,00 mm), konsentrasi Pb 15 mM menghasilkan diameter zona hambat antara 0,63-4,30 mm, dan konsentrasi Pb 30 mM menghasilkan diameter zona hambat antara 1,70-6,30 mm. Hasil analisis zona hambat menunjukkan bahwa beberapa isolat yang dihasilkan dari Telaga 2 dan Telaga 3 resisten terhadap logam Pb pada konsentrasi 6 mM dibandingkan dengan konsentrasi 15 dan 30 mM.
ENGLISH:
The ex-mining area contains pollutants in the form of lead heavy metal which can disrupt ecosystems and human health. One of the efforts to manage this problem is using the bioremediation method by utilizing bacteria. Aims of this research to identify bacteria based on morphology and see their resistance to lead heavy metal in various concentrations. Seven isolates resulted from bacterial isolation, namely T2P1a, T2P1b, T2P2, T3P1, T3P2, T3P3, and T3P4. The morphology of all isolates generally was round, brownish white, small in size, had flat edges and a raised surface. The highest number of colonies in Lake 2 and Lake 3 was produced by T2P2 and T3P2, with a colony number of 1.3 x 108 and 3.3 x 1010 cfu/mL grown on solid media without any lead heavy metal content with an incubation period of 2x24 hours at room temperature. The resistance of bacteria to various metal concentrations based on the minimum inhibitory concentration (MIC) test showed that all isolates at lead concentration 6 mM averages did not produce an inhibition zone diameter (0.00 mm), a lead concentration 15 mM resulted in an inhibition zone diameter of 0.63-4.30 mm, and a lead concentration 30 mM resulted in a zone of inhibition between 1.70-6.30 mm. The results of the inhibition zone analysis showed that some of the isolates produced from Lake 2 and Lake 3 were resistant to lead heavy metal at a concentration 6 mM compared to concentrations of 15 and 30 mM.
