Abstract
ABSTRACK
Shigella dysenteriae is a pathogenic bacteria that causes bacillary dysentery which attacks intestinal epithelial cells and causes symptoms such as bloody or slimy liquid diarrhea, abdominal pain and fever. The incidence of bacillary dysentery caused by Shigella dysenteriae is 165 million cases every year in worldwide and accounts for 29% mortality in the age group of one to four years in Indonesia. Shigella dysenteriae resistance to antibiotics has been widely reported as ampicillin, cotrimoxazole, and nalidixic acid. This resistance makes treatment of the bacillary dysentery more difficult so it is necessary to develop herbal medicine. One of the herbal plants is kaffir lime leaf (Citrus hystrix). Kaffir lime leaf is a plant that are often found in Indonesia and have antibacterial compounds such as flavonoids, alkaloids, saponins, astiri oils and tannins.
The purpose of this study was to determine the antibacterial activity of kaffir lime leaf extracts (Citrus hystrix) on the growth of Shigella dysenteriae. This research is an experimental laboratory study using paper disc diffusion methods, well diffusion and tube dilution. Extraction was carried out by the Ultrasound - Assisted Solvent Extraction method using 96% ethanol solvent. The concentrations tested on Shigella dysenteriae bacteria using extracts of kaffir lime leaves (Citrus hystrix) were 30%, 25%, 20%, 15%, 10%, and 5%. The data obtained in the form of Inhibitory Capability, MIC and MBC were analyzed with kruskall wallis.
Based on the results of the Kruskall wallis test showed a significant value of 0.00 (α <0.05) which means there is a significant influence of the antibacterial activity of kaffir lime leaf extract (Citrus hystrix) on Shigella dysenteriae. The 30% concentration of kaffir lime leaf extract (Citrus hystrix) is the best concentration in forming the inhibition zone which is 9,75 mm. Minimum Inhibitory Concentration (MIC) of Shigella dysenteriae at the lowest concentration of 5%, while at the Minimum Bactericidal Concentration (MBC) at a concentration of 20%. The conclusion of this study is the extract of kaffir lime leaves (Citrus hystrix) have antibacterial activity against Shigella dysenteriae.

Shigella dysenteriae merupakan salah satu bakteri patogen penyebab penyakit disentri basiler yang menyerang sel-sel epitel usus dan menimbulkan gejala seperti diare cair berdarah atau berlendir, nyeri perut dan demam. Angka kejadian disentri basiller yang disebabkan oleh Shigella dysenteriae sebesar 165 juta kasus tiap tahun di dunia dan menyumbang mortalitas sebanyak 29% pada kelompok umur satu sampai empat tahun di Indonesia. Resistensi Shigella dysenteriae terhadap antibiotik telah banyak di laporkan seperti ampicilin, kotrimoksazol, dan asam nalidiksat. Resistensi tersebut membuat pengobatan disenteri basiler lebih sulit sehingga diperlukan untuk mengembangkan pengobatan herbal. Salah satu tanaman herbal adalah daun jeruk purut (Citrus hystrix). Daun jeruk purut merupakan tanaman yang sering dijumpai di Indonesia dan memiliki senyawa antibakteri seperti flavonoid, alkaloid, saponin, minyak atsiri dan tanin.
Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak daun jeruk purut (Citrus hystrix) terhadap pertumbuhan bakteri Shigella dysenteriae. Penelitian ini merupakan penelitian eksperimental laboratorium dengan menggunakan metode difusi kertas cakram, difusi sumuran dan dilusi tabung. Ekstraksi dilakukan dengan metode Ultrasound – Assisted Solvent Extraction menggunakan pelarut etanol 96%. Konsentrasi yang diuji pada bakteri Shigella dysenteriae dengan menggunakan esktrak daun jeruk purut (Citrus hystrix) sebesar 30%, 25%, 20%, 15%, 10%, dan 5%. Data yang didapat berupa diameter zona hambat, KHM dan KBM dianalisis dengan kruskall wallis.
Berdasarkan hasil uji Kruskall wallis menunjukkan nilai siginifikansi sebesar 0,00 (α < 0,05) yang berarti terdapat pengaruh aktivitas antibakteri ekstrak daun jeruk purut (Citrus hystrix) secara signifikan pada bakteri Shigella dysenteriae. Konsentrasi 30% ekstrak daun jeruk purut (Citrus hystrix) merupakan konsentrasi terbaik dalam membentuk zona hambat yakni sebesar 9,75 mm. Kadar Hambat Minimum (KHM) Shigella dysenteriae pada konsentrasi terendah yaitu 5%, sedangkan pada Kadar Bunuh Minimum (KBM) pada konsentrasi 20%. Kesimpulan dari penelitian ini adalah ekstrak daun jeruk purut memiliki aktivitas antibakteri terhadap bakteri Shigella dysenteriae.
