Abstract

ABSTRACT

Cigarette smoke contains thousands of toxic chemical compounds that capable to trigger oxidative stress. One of the markers to indicate the degree of oxidative stress that occurs is Malondialdehyde (MDA). Lemon basil (Ocimum citriodorum) leaf extract is thought to have antioxidant activity as it contains a number of phenolic and non-phenolic compounds that are capable of contributing to the reduction of radical compounds and assisting the synthesis of endogenous antioxidants.

The aim of this study is to examine the effect of administering the lemon basil leaf extract to MDA levels from rats exposed by cigarette smoke. A total of 30 white male wistar strain rats were divided into six groups with different treatments. The dose of lemon basil leaf extract used is 50 mg/kgBB, 100 mg/kgBB, 200 mg/kgBB and given to the rat after exposure to cigarette smoke as much as 3 rods/day for 1 hour. After 14 days treatment, rats were terminated and MDA plasma levels were measured using by Spectrophotometer UV-Vis.

The results of the study using One Way ANOVA and Post Hoc Tukey showed there is a significant difference (p< 0.05) on the use of lemon basil leaf extract as antioxidants to MDA levels from rats that got cigarette smoke exposure with the highest MDA levels are in the negative control group and the lowest MDA levels are found in the P3 group. Based on the results can be concluded that the administration of lemon basil leaf extract is influential in lowering MDA levels in rats exposed by cigarette smoke.

ABSTRAK

Asap rokok mengandung ribuan senyawa kimia toksik yang mampu memicu stres oksidatif. Salah satu marker untuk menunjukkan derajat stres oksidatif yang terjadi adalah malondialdehida (MDA). Ekstrak etanol daun kemangi (Ocimum citriodorum) diduga memiliki aktivitas antioksidan karena mengandung sejumlah senyawa fenolik dan non fenolik yang mampu berperan dalam mereduksi senyawa radikal dan membantu sintesis antioksidan endogen.

Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak etanol daun kemangi terhadap kadar MDA tikus yang dipapar asap rokok. Sebanyak 30 ekor tikus putih wistar jantan dibagi menjadi enam kelompok dengan perlakuan yang berbeda. Dosis ekstrak daun kemangi yang digunakan adalah 50mg/kgBB, 100 mg/kgBB, dan 200mg/kgBB yang diberikan setelah paparan asap rokok sebanyak 3 batang/hari selama 1 jam. Setelah 14 hari perlakuan, tikus diterminasi dan dilakukan pengukuran kadar MDA plasma dengan Spektrofotometer UV-Vis.

Hasil penelitian menggunakan uji One Way ANOVA dan Post Hoc Tukey menunjukkan terdapat perbedaan yang signifikan (p<0,05) pada pemberian ekstrak daun kemangi terhadap kadar MDA yang dipapar asap rokok dengan rerata kadar MDA tertinggi terdapat pada kelompok kontrol negatif dan rerata kadar MDA terendah terdapat pada kelompok P3. Berdasarkan hasil penelitian dapat disimpulkan bahwa pemberian ekstrak etanol daun kemangi berpengaruh dalam menurunkan kadar MDA pada tikus yang dipapar asap rokok.
