Abstract
INDONESIA:
Koksidiosis merupakan penyakit utama penyebab kerusakan jaringan pada usus ayam. Penyakit ini disebabkan oleh tujuh spesies dari Eimeria (E. acervulina, E. brunetti, E. maxima, E. mitis, E. necatrix, E. praecox and E. tenella), dimana untuk mengatasinya memerlukan vaksin protein rekombinan. Salah satu komponen utama dalam pembuatan vaksin protein rekombinan adalah protein target. Tujuan penelitian ini adalah untuk mengetahui ekspresi protein rekombinan E. acervulina 1433 (Ea1433), E. tenella immune mapped protein 1 (EtIMP1), dan E. maxima immune mapped protein 1 (EmIMP1) sebagai protein target dalam pembuatan vaksin koksidiosis pada ayam. Jenis penelitian yang digunakan adalah deskriptif dengan sampel berupa gen EtIMP1, EmIMP1, Ea1433. Terdapat empat tahapan utama pada penelitian yaitu: sintesis gen target, penyisipan dan transformasi ke sel E. coli, ekstraksi DNA target, dan ekspresi protein rekombinan. Parameter dalam penelitian ini adalah ada tidaknya pita DNA berukuran 1180 bp (EtIMP1), 1147 bp (EmIMP1), dan 837 bp (Ea1433) serta adanya kecocokan hasil dari sekuensing. Selain itu ekspresi protein diketahui dari adanya pita protein yang dihasilkan dari western blot dengan ukuran 45 kDa (EtIMP1), 45 kDa (EmIMP1), dan 37 kDa (Ea1433). Hasil PCR dari gen EtIMP1 menunjukkan adanya pita DNA yang terbentuk redup dengan ukuran ≈ 1100 dan dua pita DNA yang sangat jelas berukuran < 750 bp. Hasil sekuensingnya tidak menunjukkan adanya kecocokan sekuens dengan spesies E. tenella. Sedangkan hasil PCR pada gen EmIMP1 tidak menunjukkan adanya pita DNA yang terbentuk. Ekspresi gen EtIMP1 dan EmIMP1 tidak dapat dilakukan karena gen target tidak dapat disintesis. Gen Ea1433 berhasil disintesis dengan ukuran 837 bp dan hasil sekuensingnya cocok. Gen Ea1433 berhasil diekspresi pada sel BL21(DE3) yang ditunjukan oleh adanya pita protein yang terbentuk dengan ukuran 37 kDa. Oleh karena itu protein target EtIMP1 dan EmIMP1 tidak dapat diekspresikan, sedangkan protein Ea1433 berhasil diekskpresikan sebagai protein target dalam pembuatan vaksin koksidiosis pada ayam.

Coccidiosis is a major disease that causes damage to tissue in the chicken intestine. This disease is caused by seven species of Eimeria (E. acervulina, E. brunetti, E. maxima, E. mitis, E. necatrix, E. praecox and E. tenella), which requires recombinant protein vaccine to overcome them. One of the main components in making recombinant protein vaccines is target proteins. The purpose of this study was to determine the expression of recombinant protein E. acervulina 1433 (Ea1433), E. tenella immune mapped protein 1 (EtIMP1), and E. maxima immune mapped protein 1 (EmIMP1) as a target protein in the production of coccidiosis vaccine in chickens. This type of research is descriptive with samples in the form of EtIMP1, EmIMP1, Ea1433 genes. There are four main stages in the research, namely: synthesis of target genes, insertion and transformation into E. coli cells, extraction of target DNA, and recombinant protein expression. The parameters in this study are the presence or absence of DNA bands measuring 1180 bp (EtIMP1), 1147 bp (EmIMP1), and 837 bp (Ea1433) and the matching results of sequencing. In addition, protein expression is known from protein bands produced from western blots of size 45 kDa (EtIMP1), 45 kDa (EmIMP1), and 37 kDa (Ea1433). PCR results from the EtIMP1 gene showed a DNA band formed dimmed with a size of ≈ 1100 and two very clear DNA bands < 750 bp in size. The results of sequencing did not show any compatibility of the sequence with E. tenella species. While the PCR results in the EmIMP1 gene did not show the presence of DNA bands formed. Expression of EtIMP1 and EmIMP1 genes cannot be done because the target gene cannot be synthesized. Ea1433 gene was synthesized with size 837 bp and the result of sequencing was matched. Ea1433 gene was successfully expressed in BL21(DE3) cells which was shown by the presence of a protein band formed in the size of 37 kDa. Therefore the EtIMP1 and EmIMP1 target proteins cannot be expressed, while the Ea1433 protein has been successfully expressed as a target protein in the production of the coccidiosis vaccine in chickens.
