Abstract
INDONESIA:
Perkembangan industri meningkatkan pencemaran yang diakibatkan oleh eksplorasi sumber daya alam secara berlebihan. Pertambangan minyak menyebabkan pencemaran logam berat, salah satunya di Desa Wonocolo, Kecamatan Kedewan, Kabupaten Bojonegoro, Jawa Timur. Kandungan tembaga (Cu) dalam tanah di lokasi pertambangan minyak tersebut sebesar 36,51-51,9 mg/kg. Menurut Peraturan Pemerintah Republik Indonesia No. 82 Tahun 2001, nilai maksimal tembaga dalam tanah sebesar 0,02 mg/L. Tectona grandis merupakan salah satu tumbuhan hiperakumulator Cu yang dapat membentuk asosiasi dengan fungi endofit. Penelitian ini bertujuan untuk menguji biosorpsi tembaga oleh fungi endofit terpilih dari daun Tectona grandis pada pertambangan minyak Desa Wonocolo, Kecamatan Kedewan, Kabupaten Bojonegoro, Jawa Timur. Penelitian ini merupakan penelitian eksperimental yang bersifat deskriptif kualitatif dan kuantitatif. Sampel daun diambil dari tiga lokasi yang berbeda dengan tiga kali ulangan pada setiap lokasi, kemudian dilakukan isolasi fungi endofit, uji toleransi, dan uji biosorpsi terhadap Cu. Parameter yang diamati berupa kemampuan toleransi dan biosorpsi serta identifikasi secara makroskopis dan mikroskopis. Uji kemampuan toleransi menggunakan CuCl2.2H2O dengan variasi konsentrasi sebesar 25, 50, 75, 100, dan 200 mg/L, sedangkan uji biosorpsi dengan variasi konsentrasi sebesar 20, 40, 60, 80, dan 100 mg/L. Analisis kandungan Cu dilakukan dengan Spektrofotometer Uv-Vis. Analisis data hasil uji kemampuan biosorpsi Cu menggunakan One Way Anova dengan selang kepercayaan 95% (α=0,05). Hasil isolasi didapatkan 1 isolat fungi endofit yang toleran terhadap konsentrasi Cu 200 mg/L yang memiliki indeks toleransi sebesar 0,5625 dan termasuk dalam genus Aspergillus. Kemampuan biosorpsi isolat tersebut pada konsentrasi Cu 20, 40, 60, 80, dan 100 mg/L secara berturut-turut sebesar 99.95%, 99.36%, 99.11%, 98.52%, dan 97.29%.

Industrial development increases pollution caused by exploration of natural resources excessively. Oil mining has caused heavy metal pollution, one of which is in Wonocolo Village, Kedewan District, Bojonegoro Regency, East Java. The copper content in the soil at the oil mining is 36.51-51.9 mg/kg. According to Republic of Indonesian Government Regulation No. 82 of 2001, the maximum value of copper in the soil is 0,02 mg/L. Tectona grandis is one of the copper hyperaccumulator plants that can form associations with endophytic fungi. This study aims to test the biosorption of copper by selected endophytic fungi from Tectona grandis leaves in the oil mining of Wonocolo Village, Kedewan District, Bojonegoro Regency, East Java. This research is an experimental research using descriptive qualitative and quantitative. Leaf samples were taken from three different locations with three replications at each location, then isolation of endophytic fungi, tolerance tests, and biosorption tests o copper. The parameters observed were tolerance and biosorption ability as well as macroscopic and microscopic identification. Tolerance ability test use CuCl2.2H2O with concentration 25, 50, 75, 100, and 200 mg/L and biosorption tests with concentration 20, 40, 60, 80, and 100 mg/L. Analysis of copper content was performed with a Uv-Vis Spectrophotometer. Data analysis of copper biosorption test results using One Way Anova with a confidence interval of 95% (α = 0.05). Isolation results obtained 1 endophytic fungi isolate which is tolerant to the concentration of copper 200 mg/L which has a tolerance index of 0.5625 and belongs to the genus Aspergillus. The biosorption ability of these isolate at concentrations of copper 20, 40, 60, 80, and 100 mg/L were 99.95%, 99.36%, 99.11%, 98.52%, and 97.29% respectively.
