Abstract
ABSTRACT
Heavy metal contamination has become a serious problem, the level of concentration of heavy metals can cause toxicity to biological organisms and natural imbalances. Wonocolo Village is one of the villages in Bojonegoro that produces oil traditionally. Copper or (Cu) is one of the several heavy metals that accumulates in oil-contaminated soils. Preliminary tests conducted at the petroleum mining in Wonocolo Village ranged from 36.51 to 61.9 mg / kg. The range of levels is included in the dangerous category, according to the Energy and Mineral Resources Regulation of 2006 the TCLP (Toxicity Characteristic Leaching Procedure) standard for Cu is 10 mg / kg. Based on Pratiwi's research (2019), that 2 isolates from the endophytic fungi of the Tridax procumbens L. plant have the potential to absorb heavy metal copper (Cu) with biosorption efficiency of 89.68% and 85.44% in 200 ppm Cu. The level of efficiency in the absorption of heavy metals in the biosorption process involves several factors, one of which is the pH value, optimum temperature and heavy metal concentration levels. This study aims to determine the tolerance index, pH and concentration of heavy metal concentrations to increase the biosorption efficiency and to determine the ability of the two isolates to absorb Cu metal from several concentrations. This research is an experimental study with quantitative description research design. The fungi samples used were 2 fungi isolates from Tridax procumbens roots. The parameters observed were biosorption ability with variations in pH (3,7,9) and concentration of Cu (200,300,400) ppm. Analysis of Cu content was performed with a UV-Vis spectrophotometer. Quantitative test results were analyzed using ANAVA. If it is significantly different, it is tested further with the Post Hoc Tukey 95% test using the SPSS program. The results obtained that the isolate I1 had the highest tolerance level of 68.40% under conditions of pH 3 (acidic) and 200 ppm. While the isolate I2 code has the highest Cu biosorption ability at pH 3 (acidic) with a final Cu level of 6.54 ppm with a biosorption percentage of 97.24% compared to isolate I1 at a concentration of 400 ppm.

Kontaminasi logam berat telah menjadi masalah serius, tingkat konsentrasi logam berat dapat menyebabkan toksisitas terhadap organisme biologis dan ketidakseimbangan alam. Desa Wonocolo merupakan salah satu desa di Bojonegoro yang memproduksi minyak bumi secara tradisional. Tembaga atau (Cu) adalah salah satu dari beberapa logam berat yang terakumulasi pada tanah tercemar pertambangan minyak bumi. Uji pendahuluan yang telah dilakukan di pertambangan minyak bumi di Desa Wonocolo berkisar antara 36.51-61.9 mg/kg. Kisaran kadar tersebut termasuk dalam kategori berbahaya, menurut Peraturan Energi dan Sumber Daya Mineral tahun 2006 kadar baku TCLP (Toxicity Characteristic Leaching Procedure) untuk Cu adalah 10 mg/kg. Berdasarkan penelitian Pratiwi (2019), bahwa 2 isolat dari fungi endofit tanaman Tridax procumbens L. berpotensi mengabsorbsi logam berat tembaga (Cu) dengan efisiensi biosorpsi 89.68% dan 85.44% dalam 200 ppm Cu. Tingkat efesiensi dalam penyerapan logam berat pada proses biosorpsi melibatkan beberapa faktor, salah satunya adalah nilai pH, suhu optimum dan kadar konsentrasi logam berat. Penelitian ini bertujuan untuk mengetahui indeks toleransi, pH dan kadar konsetrasi logam berat untuk meningkatkan nilai efisiensi biosorpsi dan mengetahui kemampuan kedua isolat dalam mengabsorpsi logam Cu dari beberapa konsentrasi. Penelitian ini merupakan penelitian eksperimental dengan rancangan penelitian deskripsi kuantitatif. Sampel fungi yang digunakan yaitu 2 isolat fungi dari akar Tridax procumbens. Parameter yang diamati adalah keampuan biosorpsi dengan variasi pH (3,7,9) dan konsentrasi Cu (200,300,400) ppm. Analisa kandungan Cu dilakukan dengan Spektrofotometer UV-Vis. Hasil uji kuantitatif dianalisis menggunakan ANAVA. Jika berbeda nyata diuji lanjut dengan uji Post Hoc Tukey 95% menggunakan program SPSS. Hasil yang didapatkan bahwa isolat I1 memiliki tingkat toleransi tertinggi 68.40 % pada kondisi pH 3 (asam) dan 200 ppm Cu. Sedangkan kode I2 memiliki kemapuan penghilangan Cu paling tinggi pada kondisi pH 3(asam) dengan kadar Cu akhir sebesar 6.54 ppm dengan prosentase biosorpsi sebesar 97.24% dibandingkan dengan isolat I1 pada konsentrasi 400 ppm.
