Abstract
INDONESIA:
Tanaman anting-anting (Acalypha indica Linn) merupakan tanaman liar yang berpotensi dijadikan sebagai obat. Acalypha indica Linn diketahui mengandung senyawa golongan alkaloid isokuinolin berberin. Penelitian ini dilakukan untuk mengetahui kondisi optimal pemisahan dan mengindentifikasi senyawa berberin dalam tanaman anting-anting menggunakan KCKT untuk kemudian ditentukan kadarnya.
Sampel dalam penelitian ini di ekstraksi menggunakan pelarut etil asetat dengan metode ekstraksi ultrasonik selama 20 menit pada frekuensi 42 KHz. Optimasi parameter sistem KCKT yang dilakukan yakni pada laju alir 1,0 mL/min dengan jenis fase gerak metanol : asam format (0,1%) dan asetonitril : asam format (0,1%) pada beberapa variase sistem elusi. Instrumen KCKT yang digunakan UFLC SPD-20A, prominence UV/Vis detector, kolom C-18 4,6 x 250 mm, 5 μm dengan metode gradien pada panjang gelombang 345 nm.
Kondisi optimum yang diperoleh berada pada fase gerak metanol : asam format (0,1%) dengan sistem elusi 0-10 menit 80% asam format (0,1%), 10-20 menit 50% asam format (0,1%), 20-25 menit 50% asam format (0,1%), 25-30 menit 80% asam format (0,1%). Identifikasi menggunakan metode adisi menghasilkan kenaikan intensitas pada puncak senyawa berberin berada pada waktu retensi 15,492. Hasil analisis kuantitatif menunjukkan kadar senyawa berberin dalam ekstrak etil asetat tanaman anting-anting sebesar 33,578 ppm.

Anting-anting plants (Acalypha indica Linn) is a wild plant that has the potential to be used as medicine. Acalypha indica Linn is known to contain a compound of isokuinolin alkaloids of berberine. This research was conducted to determine the optimal conditions of separation and to identify berberine compounds in anting-anting plants using HPLC to determine the levels.
The sample in this research was extracted using ethyl acetate solvent with ultrasonic extraction method for 20 minutes at a frequency of 42 KHz. The optimization of the HPLC system parameters was carried out at a flow rate of 1.0 mL / min and the type of mobile phase of methanol : formic acid (0.1%) and acetonitrile : formic acid (0.1%) with several variations of the elution system. HPLC instrument used UFLC SPD-20A, UV/Vis detector, C-18 column 4.6 x 250 mm, 5 μm with gradient method at a wavelength of 345 nm.
The optimum conditions obtained were in the mobile phase of methanol : formic acid (0.1%) with an elution system 0-10 minutes 80% formic acid (0,1%), 10-20 minutes 50% formic acid (0.1%) , 20-25 minutes 50% formic acid (0.1%), 25-30 minutes 80% formic acid (0.1%). Identification using the addition method shows that the berberine compound is at a retention time of 15.492 minutes. The results of quantitative analysis showed the levels of berberine compounds in the anting-anting plants ethyl acetate extract amounted to 33.578 ppm.
