Abstract
INDONESIA :
Tanaman rumput bambu mengandung senyawa tanin yang berpotensi sebagai antikanker. Perkembangan penelitian menunjukkan bahwa potensi senyawa antikanker dapat ditingkatkan melalui pengembanan pada zeolit sebagai sistem penghantar obat. Penelitian ini bertujuan untuk mengetahui aktivitas antikanker ekstrak dan isolat tanin rumput bambu yang diembankan pada zeolit NaX terhadap sel kanker payudara T47D.
Ekstraksi senyawa aktif rumput bambu dilakukan menggunakan pelarut aseton : air (7:3) dan asam askorbat 10 mM. Ekstrak yang didapat dipartisi menggunakan kloroform dan etil asetat. Hasil maserasi dan partisi diuji fitokimia untuk mengetahui kandungan tanin secara kualitatif. Isolasi senyawa tanin dilakukan dengan metode KLTP menggunakan eluen n-butanol : asam asetat : air (4:1:5). Ekstrak dan isolat tanin rumput bambu diembankan pada zeolit NaX dengan metode impregnasi kering dengan perbandingan 5:10. Hasil pengembanan dikarakterisasi menggunakan FTIR dan diuji aktivitas antikanker pada sel kanker payudara T47D dengan metode MTT.
Hasil uji fitokimia menunjukkan ekstrak dan fraksi air rumput bambu mengandung senyawa tanin. Munculnya beberapa pita serapan baru pada hasil karakterisasi FTIR sampel hasil pengembanan menunjukkan ekstrak dan isolat tanin rumput bambu telah teremban. Nilai IC50 ekstrak, isolat tanin, zeolit NaX, hasil pengembanan ekstrak dan isolat tanin rumput bambu secara berturut-turut yaitu 107,240; 284,936; 751,380; 423,837; dan 560,262 µg/mL. Ekstrak dan isolat tanin rumput bambu yang diembankan pda zeolit NaX dalam penelitian ini kurang berpotensi aktif sebagai antikanker terhadap sel kanker payudara T47D.

Bamboo grass (Lophaterum gracile B.) contains tannin compounds that has the potential as anticancer. The development of research shows that the potential of anticancer compounds can be increased through loading on zeolites as a drug delivery system. This study aimed to determine the anticancer activity of extracts and tannin isolate of bamboo grass which were loaded on NaX zeolite against breast cancer cells T47D.
Extraction of active compounds of bamboo grass was carried out using acetone: water (7:3) and 10 mM ascorbic acid. The extract obtained was partitioned using chloroform and ethyl acetate. The results of maceration and partition were phytochemicals tested to determine the tannin content qualitatively. Isolation of tannin compounds was carried out by the KLTP method using eluent n-butanol: acetic acid: water (4: 1: 5). Extracts and tannin isolate of bamboo grass were loaded on NaX zeolite using dry impregnation method with ratio of 5:10. The results were characterized using FTIR and tested for anticancer activity against breast cancer cells T47D by MTT method.
Phytochemical test results showed that extracts and water fractions of bamboo grass containing tannin compounds. The appearence of several new absorption bands in the results of FTIR characterization of the samples showed that extracts and tannin isolate of bamboo grass had been loaded. The IC50 values of extracts, tannin isolate, NaX zeolite, the results of loading bamboo grass extracts and tannin isolates respectively are: 107,240; 284,936; 751,380; 423,837; and 560,262 µg/mL. Extract and tannin isolate of bamboo grass which were loaded on NaX zeolite in this study were less potential to be active as anticancer against breast cancer cells T47D.
