Abstract
INDONESIA:
Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan senyawa steroid yang dikhususkan pada tumbuhan Hydrilla verticillata beserta identifikasinya. Penelitian dilakukan dengan preparasi sampel Hydrilla verticillata yang dilanjutkan dengan ekstraksi menggunakan metanol. Ekstrak pekat metanol kemudian dihidrolisis menggunakan HCl dan dilanjutkan partisi menggunakan n-heksana. Hasil partisi yang diperoleh kemudian diuji fitokimia senyawa steroid dan dipisahkan senyawanya menggunakan KLT. Serta diidentifikasi menggunakan spektrofotometer UV-Vis, FTIR, dan LC-MS/MS. Hasil penelitian menunjukkan ekstraksi maserasi menghasilkan rendemen 5,14%, dan rendemen dari partisi n-heksan sebesar 47,95%. Pemisahan steroid menggunakan Kromatografi Lapis Tipis Analitik (KLTA) menunjukkan bahwa variasi eluen terbaik adalah n-heksana: etil asetat (4:1) dengan 12 spot. Pemisahan Kromatografi Lapis Tipis Preapratif (KLTP) menghasilkan 19 spot. Isolat hasil KLTP diuji antioksidan menghasilkan EC50 5,109 ppm. Identifikasi senyawa steroid menggunakan UV-Vis menunjukkan bahwa panjang gelombang maksimum sebesar 203,9 nm dan 276 nm, sedangkan untuk identifikasi isolat steroid menggunakan spektrofotometer FTIR yang menunjukkan gugus fungsi O-H, geminal dimetil, C=O, C=C, C-OH sekunder dan =C-H (alkena) yang diduga merupakan senyawa steroid. Hasil pengujian LC-MS/MS menunjukkan adanya 1 senyawa jenis steroid yaitu β-sitosterol.
ENGLISH:
The objectives of this study were to identify and determine the antioxidant activity of steroid derivative isolated from Hydrilla verticillata. The research was initiated with sample preparation of Hydrilla verticillata and followed by extraction using methanol. Hydrolization of concentrated methanol extract with HCl preceded compounds partition using n-hexane. The results of partition process were tested phytochemically for steroid compound which was separated through thin layer chromatography. The obtained steroid derivative was identified using UV-Vis spectrophotometer, FTIR and LC-MS/MS. The result of the study showed that extraction through maceration produced 5.14% yield, whereas product yield of partition using n-hexane was around 47.95%. Steroid separation through analytical thin layer chromatography (ATLC) revealed that the best eluant variation was n-hexane:acetyl acid (4:1) which resulted in 12 spots. Preparative thin layer chromatography (PTLC) yielded 19 spots. Isolate obtained from preparative TLC was assessed for its antioxidant activity and exhibited EC50 of 5.109 ppm. Identification of steroid compound using UV-Vis spectrophotometer displayed maximum wavelength at 203.9 nm and 276 nm, while the identification of steroid compound with FTIR indicated the presence O-H functional group, geminal dimethyl, C=O, C=C, secondary C-OH and =C-H (alkene) which are presumably of a steroid compound. The result of LC-MS/MS showed the presence of 1 compound of steroid derivative, β-sitosterol.
