Abstract
INDONESIA :
Sel otak merupakan sel yang rentan mengalami kerusakan dalam kondisi in vitro karena memiliki kelangsungan hidup yang singkat dan selektif mengenai lingkungan tempat tumbuhnya. Oleh sebab itu, viabilitas dan nilai Population Doubling Time (PDT) atau waktu proliferasi sel otak dapat menurun. Ekstrak propolis diduga mampu melindungi sel otak dari kerusakan dan mampu meningkatkan viabilitas serta mempercepat nilai PDT secara in vitro. Hal ini dikarenakan ekstrak propolis memiliki senyawa aktif yang berpotensi sebagai neuroprotektif, antioksidan, dan stimulator pertumbuhan sel. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak propolis terhadap viabilitas dan proliferasi sel otak tikus (Rattus norvegicus) secara in vitro. Penelitian ini merupakan penelitian eksperimental yang menggunakan rancangan acak lengkap (RAL) dengan 6 perlakuan dan 4 ulangan. Perlakuan yang digunakan adalah P1 (kontrol) dan perlakuan pemberian ekstrak propolis yaitu P2 (20 µg/mL), P3 (40 µg/mL), P4 (60 µg/mL), P5 (80 µg/mL), dan P6 (100 µg/mL). Sampel yang digunakan adalah sel otak tikus (Rattus norvegicus) umur 3-4 hari yang dikultur dalam media DMEM 10% FBS dan diinkubasi dalam inkubator bersuhu 37oC dengan CO2 5% selama 10 hari. Ekstraksi propolis menggunakan metode maserasi dengan pelarut etanol 96%. Data hasil penelitian berupa nilai viabilitas (%) dan nilai PDT (hari) sel otak tikus (Rattus norvegicus) dianalisis menggunakan one way ANOVA (α=0.05). Berdasarkan hasil analisis menggunakan one way ANOVA, dapat disimpulkan bahwa tidak ada pengaruh pemberian ekstrak propolis terhadap viabilitas dan nilai PDT sel otak tikus (p<0.05). Akan tetapi, rata-rata nilai viabilitas tertinggi (90.3 ± 3.09%) dan nilai PDT tercepat (2.88 ± 0.66 hari) dicapai oleh sel otak yang dikultur dengan pemberian perlakuan P5 (80 µg/mL).

Brain cells are susceptible cell in in vitro conditions because of their short life span and highly specialized and choosy about their environment, so cell viability and Population Doubling Time (PDT) value or time for brain cells to proliferation has the potential to decrease. Propolis extract is expected to be able to protect brain cells from damage and able to increase viability and accelerate PDT values because it has active compounds which are potentially to be neuroprotective, antioxidants, growth cell stimulator. The purpose of this research were to determine the effect of propolis extract on the viability and proliferation of rat (Rattus norvegicus) brain cells in vitro. This research was an experimental study which was using Completely Randomized Design (CRD) with 6 treatments and 4 replications. The treatments were P1 (control) and treatments with additions of propolis extract i.e. P2 (20 µg/mL), P3 (40 µg/mL), P4 (60 µg/mL), P5 (80 µg/mL), dan P6 (100 µg/mL). Sample of this research was 3-4 day rat (Rattus norvegicus) brain cells which were cultured in DMEM 10% FBS and incubated in a 37oC incubator with 5% CO2 for 10 days. Propolis extraction was using maceration method with 96% ethanol. Data of this research were viability (%) and PDT values (days) of rat (Rattus norvegicus) brain cells which were the analyzed using one way ANOVA (α=0.05). Based on one way ANOVA, it was concluded that additions of propolis extract did not affect brain cell viability and PDT values (p <0.05). However, the highest average of viability (90.3 ± 3.09%) and the fastest PDT value (2.88 ± 0.66 days) achieved by brain cells cultured by addition of P5 (80 µg/mL).
