Abstract
INDONESIA:
Khamir merupakan mikroorganisme uniseluler eukariotik yang memiliki berbagai macam potensi khususnya dalam fermentasi karbohidrat. Eksplorasi khamir pada bunga pisang klutuk (Musa balbisiana) perlu dilakukan karena merupakan sumber karbohidrat tertinggi diantara bunga lainnya. Tujuan dari penelitian ini adalah untuk mengetahui jenis khamir yang berasosiasi dengan bunga pisang klutuk (Musa balbisiana) berdasarkan karakter morfologi dan molekuler serta potensinya dalam dalam fermentasi karbohidrat. Penelitian ini terdiri dari beberapa tahap, yaitu: isolasi, purifikasi, identifikasi khamir secara makroskopik dan mikroskopik, uji fermentasi karbohidrat dan gula 50%. Isolate khamir dengan kemampuan fermentasi terbaik dipilih untuk identifikasi molekuler dengan menggunakan primer ITS1 dan ITS4. Hasil isolasi dan identifikasi didapatkan 7 (YI-1, YI-2, YI-3, YI-4, YI-5, YI-6 dan YI-7) isolat khamir berdasarkan karakter morfologi dimana semua jenis isolate mempunyai kemiripan dengan anggota kelas Ascomycetes. Berdasarkan uji fermentasi karbohidrat (fruktosa, glukosa, dan galaktosa) didapat tiga isolat terbaik yaitu isolat YI-4, YI-6, dan YI-7. Hasil analisa genetika diketahui bahwa isolat YI-4 mempunyai kemiripan dengan Saccharomyces cerevisiae strain HBUM07151 (99%), isolat YI-6 dengan Saccharomyces cerevisiae isolate E1 (98%), dan isolat YI-7 dengan Saccharomyces cerevisiae isolate B-WHX-12-48 (99%).

Yeast is eukaryotic unicellular microorganism which has potentials in it, especially in carbohydrate fermentation. Yeast exploration on the klutuk banana flower (Musa balbisiana) needs to be done due to the highest source of carbohydrates among other flowers. The purpose of the research is to determine the type of yeast that is associated with klutuk banana flower (Musa balbisiana) based on morphological and molecular characters and the potentials in carbohydrate fermentation. The research consists of several stages, namely: isolation, purification, identification of yeast macroscopically and microscopically, carbohydrate fermentation test and 50% sugar. Yeast isolate with the best fermentation ability is chosen for molecular identification using primary ITS1 and ITS4. The isolation and identification results obtained 7 (YI-1, YI-2, YI-3, YI-4, YI-5, YI-6 and YI-7) of yeast isolates based on morphological characters in which all isolates have similarities with members of the Ascomycetes class. Based on carbohydrate fermentation tests (fructose, glucose, and galactose) were obtained three best isolates, namely YI-4, YI-6, and YI-7 isolates. Genetic analysis revealed that YI-4 isolate had similarities with Saccharomyces cerevisiae strain HBUM 07151 was (99%), YI-6 isolate with Saccharomyces cerevisiae isolate E1 was (98%), and YI-7 isolate with Saccharomyces cerevisiae isolate B-WHX-12-48 was (99%).
