Abstract
INDONESIA:
Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh 2,4-D dan BA terhadap induksi kalus Bidara Upas (Merremia Mammosa). Bahan yang digunakan yaitu media MS, zat pengatur tumbuh, organ daun sebagai eksplan kalus, kalus tersebut digunakan untuk memperbanyak metabolit sekunder yang digunakan sebagai antikanker, antidiabetes, antiinflamasi, peningkatan kandungan metabolit sekunder ini diperoleh melalui kultur kalus kompak dengan penambahan 2,4-D dan BA sehingga produksi meningkat. Penelitian ini bersifat eksperimental menggunakan rancangan acak lengkap(RAL) dengan 20 perlakuan dan 3 ulangan. Perlakuan dalam penelitian ini ada dua factor yaitu konsentrasi 2,4-D meliputi 0 mg/l, 0.5 mg/l, 1 mg/l, 1.5 mg/l, 2 mg/l dan konsentrasi BA 0 mg/l, 0.5 mg/l, 1 mg/l, 1,5 mg/l. Data dianalisis dengan uji ANAVA Two Way α= 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan dengan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5 % derta uji regresi korelasi. Hasil dari penelitian kalus menunjukkan adanya pengaruh konsentrasi 2,4-D dan BA terhadap induksi kalus Bidara Upas (Merremia Mammosa ) yang optimal pada kombinasi 1 mg/l 2,4-D dan 1 mg/l BA hari muncul kalus hari ke-12 HST dengan persentase tumbuh kalus pada eksplan sebesar 92,66 % dan berat kalus sebesar 0,387 gr.
ENGLISH:
The research aims at determining the influence of growth regulator substance of 2,4-D and BA against callus induction of Bidara Upas (Merremia Mammosa). The material is MS media, growth regulator substance, leaf organs as callus explants, the callus is used to multiply secondary metabolites that are used as anticancer, anti-diabetic, anti-inflammatory, increased content of secondary metabolites are obtained through unified callus culture with the addition of 2,4-D and BA , it make the production increases. The research was experimental by using a completely randomized design (CRD) with 20 treatments and 3 replications. The treatments of the research were two factors, 2,4-D concentration included 0 mg/l, 0.5 mg / l, 1 mg/l, 1.5 mg/l, 2 mg/l and the concentration of BA included 0 mg/l, 0.5 mg/l, 1 mg/l, 1.5 mg/l. Data were analyzed by Two Way ANAVA test α = 5%. If there is a significant difference, it will be followed by the Duncan Multiple Range Test (DMRT) with a significant level of 5% and a correlation regression test. The results of the callus research showed the influence of 2,4-D and BA concentrations on the callus induction of optimal Bidara Upas (Merremia Mammosa) in a combination of 1 mg/l 2,4-D and 1 mg/l BA on the 12th (HST) day of callus day with the percentage of callus growth in explants was 92.66% and callus weight was 0.387 gr.
