Abstract
INDONESIA:
Marsilea crenata C Presl merupakan sumber senyawa fitoestrogen serta diduga dapat menghambat proses neurodegeneratif. Tujuan dilakukannya penelitian ini untuk mengetahui aktivitas antineuroinflamasi ekstrak etanol 96% daun M. crenata secara in silico dan in vitro. Senyawa hasil metabolite profilling UPLC QToF MS/MS ekstrak etanol 96% daun M. crenata dilakukan skrining menggunakan SwissADME dengan parameter TPSA dan BBB permeant. Senyawa inklusi dilakukan penambatan molekuler terhadap protein 3OLS menggunakan Autodock Vina. Sel mikroglia HMC3 diinduksi dengan IFN γ sebagai model neuroinflamasi selama 48 jam dan kemudian dibagi kedalam kelompok kontrol positif , kontrol negatif serta kelompok sel yang diberikan perlakuan dosis 62,5 ppm; 125 ppm; 250 ppm ekstrak etanol 96% M. crenata dan kemudian diamati menggunakan metode ICC dengan bantuan CLSM. Ekstrak etanol 96% daun M. crenata mengandung fitoestrogen dengan adanya 3 senyawa yang berinteraksi agonis 17β estradiol terhadap 3OLS diantaranya Prochlorperazine; 1-methyl -2-[(4-methylpiperazin-1-yl)methyl] benzimidaol- 5-amine; 11-Aminododecanoic acid serta hasil uji LSD menunjukkan terdapat perbedaan signifikan pada kelompok 250 ppm perlakuan dosis terhadap kontrol negatif dengan nilai p < 0,05 dengan ED50 3,795 ppm. Hasil penelitian menunjukkan ekstrak etanol 96% daun M. crenata memiliki senyawa fitoestrogenik sebagai agen antineuroinflamasi yang dapat mengurangi ekspresi MHC II sel mikroglia HMC3 dengan menggunakan dosis 250 ppm

Marsilea crenata C Presl is a source of phytoestrogen compounds and predict to inhibit neurodegenerative processes. The purpose of this study was to determine the antineuroinflammation activity of 96% ethanol extract M. crenata leaves in silico and in vitro. Metabolite profilling compounds of 96% ethanol extract of M. crenata leaves from UPLC QToF MS / MS were screened using SwissADME with TPSA and BBB permeant parameters. Inclusion compounds were carried out molecular docking of 3OLS proteins using Autodock Vina. HMC3 microglia cells were induced with IFN γ as a neuroinflammatory model for 48 hours and then divided into positive control groups, negative controls and cell groups given a dose of 62.5 ppm; 125 ppm; 250 ppm 96% ethanol extract M. crenata and then observed using the ICC method with CLSM. 96% ethanol extract of M. crenata leaves containing phytoestrogens in the presence of 3 compounds that interact 17β estradiol agonists against 3OLS including Prochlorperazine; 1-methyl -2 - [(4-methylpiperazin-1-yl) methyl] benzimidaol-5-amine; 11-Aminododecanoic acid and LSD test results showed that there was a significant difference in the 250 ppm group in the treatment of doses against negative controls with a value of p <0.05 with ED50 value 3.795 ppm. The results showed 96% ethanol extract of M. crenata leaves had phytoestrogenic compounds as antineuroinflammation agents that can reduce the expression of MHC II HMC3 microglia cells using a dose of 250 ppm
