Pengaruh paparan audio Murattal Al-Fatihah terhadap sel kanker HeLa secara in vitro

Kanker adalah penyakit yang ditandai dengan mekanisme sel tidak normal dan tidak terkontrol. Pengembangan pengobatan alternatif kanker saat ini masih perlu dilakukan, salah satunya dengan terapi musik. Terapi musik menggunakan vibrasi suara untuk meningkatkan kesembuhan. Pembacaan Al-Quran dengan tartil menghasilkan nada yang indah. Al-Fatihah merupakan bagian dalam Al-Quran yang sering dibaca umat Islam dan dijadikan doa untuk kesembuhan. Tujuan penelitian ini untuk mengetahui pengaruh sitotoksik murattal Al-Fatihah terhadap sel vero, sel HeLa dan kombinasinya dengan cisplatin serta pengaruhnya terhadap apoptosis dan siklus sel HeLa. Murattal Al-Fatihah diuji sitotoksik dengan metode MTT Assay terhadap sel vero, sel HeLa, dan kombinasi dengan cisplatin 10 dan 20 ppm. Setelah diketahui viabilitas, uji lanjutan dilakukan dengan metode flowcytometry untuk mengetahui siklus sel dan apoptosis. Hasil viabilitas paparan murattal terhadap sel vero, sel HeLa, sel HeLa + cisp 10 ppm dan kontrol, sel HeLa + Cisp 20 ppm dan kontrol berturut-turut yaitu 75,97%; 80,14%; 64,32%; 69,86%; 43 %; dan 43,16%. Hasil flowcytometry pada sel vero terdapat penghambatan pada fase G0-1 dan S tetapi tidak menginduksi apoptosis. Pada treatment paparan murattal tanpa cisplatin maupun dengan kombinasi cisplatin pada sel HeLa terdapat penghambatan di fase G2-M dan induksi apoptosis pada fase M5. Suara Murattal Al-Fatihah dapat memberikan pengaruh sitotoksik terhadap sel HeLa serta memberikan efek sinergi terhadap cisplatin sehingga terapi paparan murattal bisa direkomendasikan untuk terapi pendukung dalam pengobatan penyakit kanker (supportive therapy).

Cancer is a disease characterized by abnormal and uncontrolled cell mechanisms. The development of alternative cancer treatments is still needed, one of them is music therapy. Music therapy uses sound vibrations to improve healing. Al-Quran recitation with tartil produces beautiful tones. Al-Fatihah is a part of the Qur'an that is often read by Muslims and used as a prayer for healing. The purpose of this study was to determine the cytotoxic effect of murattal Al-Fatihah on vero cells, HeLa cells and their combination with cisplatin and its effect on apoptosis and HeLa cell cycle. Murattal Al-Fatihah was cytotoxically tested by the MTT Assay method on vero cells, HeLa cells, and its combination with cisplatin 10 and 20 ppm. After knowing viability, further tests were carried out using the flowcytometry method to determine cell cycles and apoptosis. Results of viability of murattal exposure to vero, HeLa, HeLa + Cisp 10 ppm and control, HeLa + Cisp 20 ppm and controls respectively were 75.97%; 80.14%; 64.32%; 69.86%; 43%; and 43.16%. The results of flowcytometry in vero cells were inhibited in the G0-1 and S phases but did not induce apoptosis. In the treatment of murattal exposure without cisplatin or with cisplatin combination in HeLa cells, there was an inhibition in the G2-M phase and induction of apoptosis in the M5 phase. Murattal Al-Fatihah can provide cytotoxic effects on HeLa cells and provide a synergistic effect on cisplatin, so that murattal Al-Fatihah exposure therapy can be recommended for supporting therapy in the treatment of cancer.
