Abstract
INDONESIA:
Senyawa dari tumbuhan berkhasiat serupa estrogen alami dikenal dengan istilah fitoestrogen dapat diperoleh dari tumbuhan semanggi (Marsilea crenata). Senyawa fitoestrogen mampu menggantikan peran estrogen dalam berikatan langsung dengan reseptor estrogen dalam proses remodeling tulang. Penelitian ini bertujuan untuk mengetahui aktivitas ekstrak etanol 96% daun Marsilea crenata terhadap peningkatan jumlah sel osteoblas tulang trabekular vertebra mencit yang diinduksi deksametason. Penelitian dilakukan pada 36 ekor mencit jantan sehat usia 5-6 bulan dikelompokkan secara random menjadi 6 kelompok. Kelompok kontrol positif dengan pemberian suspensi alendronat 0.026 mg/20gBB, kelompok kontrol negatif dengan pemberian suspensi CMC- Na, kelompok pemberian ekstrak etanol 96% daun Marsilea crenata dengan empat variasi dosis yaitu 1.2 mg, 2.4 mg, 4.8 mg dan 9.6 mg/ 20 g BB mencit per hari. Perlakuan pada setiap kelompok ini dilakukan selama 28 hari setelah mencit mengalami osteoporosis. Pengamatan terhadap peningkatan jumlah sel steoblas tulang trabekular vertebra mencit secara histomorfometri dengan pewarnaan Hematoxylin Eosin. Peningkatan jumlah sel osteoblas tulang trabekular vertebra diamati menggunakan mikroskop dengan perbesaran 100x. Hasil pengamatan menunjukkan bahwa ekstrak etanol 96% daun Marsilea crenata dapat meningkatkan jumlah sel osteoblas tulang trabekular vertebra scara signifikan dengan dosis efektif (ED50) sebesar 2.878 mg/20 g BB mencit sehingga dapat disimpulkan bahwa ekstrak etanol 96% daun Marsilea crenata memiliki aktivitas antiosteoporosis ditunjukkan dengan peningkatan jumlah sel osteoblas tulang trabekular vertebra pada mencit.
ENGLISH:
The compounds derived from plants that have a similar properties to natural estrogen were known by phytoestrogens can be obtained from Marsilea crenata. Phythoestrogen can be replaced the role of estrogen to bind directly with estrogen receptor for bone remodelling process. This study conducted to determine the activity of 96% ethanol extract of Marsilea crenata leaves to increase the number of vertebral trabecular bone osteoblasts induced by dexamethasone. The study was conducted on 36 healthy male mice aged 5-6 months randomly grouped into 6 groups. Positive control group with alendronate 0.026 mg / 20 g BB suspension, negative control group with CMC-Na suspension, group giving 96% ethanol extract of Marsilea crenata leaves with four dose variations they were 1.2 mg, 2.4 mg, 4.8 mg and 9.6 mg / 20 g BB mice per day. Each treatment of these groups had been done for 28 days after mice had an osteoporosis. The increasing of vertebrae trabecular osteoblast number of male mice was observed using optical microscope with 100x zoom in after Histomorphometry and Hematoxylin and Eosin (HE) staining process. The result showed that the 96% ethanol extract of leaves of Marsilea crenata can significantly increase the number of vertebral trabecular bone osteoblasts with effective doses (ED50) of 2,878 mg / 20 g BB mice so it can be concluded that the 96% ethanol extract of Marsilea crenata has an antiosteoporosis activity which showed with an increasing the number of vertebral trabecular bone osteoblasts in mice.
