Abstract
INDONESIA:
Staphylococcus aureus (S aureus) merupakan salah satu bakteri patogen di Indonesia yang menjadi salah satu penyebab infeksi jerawat. Getah Jatropha curcas Linn (J curcas) diidentifikasi mengandung senyawa golongan flavonoid, tannin, terpenoid, saponin dan alkaloid yang mempunyai efek antibakteri. Penelitian ini bertujuan untuk mengetahui kandungan senyawa dan aktivitas getah J curcas dalam menghambat pertumbuhan bakteri S aureus. Getah J curcas dikeringkan mengunakan oven dengan suhu 40°C selama 7x24 jam. Getah kering yang diperoleh kemudian dilakukan skrining fitokimia menggunakan uji KLT yang dilanjutkan dengan TLC-Visualizer dan selanjutnya getah kering dilakukan analisis senyawa aktif menggunakan Ultra Performance Liquid Chromatography - Mass Spectrometry (UPLC-MS). Uji aktivitas antibakteri dilakukan dengan metode difusi cakram Kirby-bauer menggunakan 4 variasi konsentrasi (25 ppm, 50 ppm, 75 ppm, dan 100 ppm) dan uji Kadar Hambat Minimum (KHM) dilakukan dengan metode pengenceran bertingkat (serial dilusi) atau metode turbidimetri. Kontrol positif menggunakan Amoxicillin 100 ppm sedangkan kontrol negatif menggunakan water for injection (WFI). Getah J curcas mempunyai bentuk kristal amorf yang larut dalam air, etanol dan metanol. Hasil analisis UPLC-MS menunjukkan getah J curcas mengandung senyawa pyridine yang merupakan golongan dari alkaloid dan beberapa senyawa penyusun lainnya seperti turunan alkohol, turunan asam karboksilat dan cyclo (cincin tertutup). Aktivitas antibakteri getah J curcas terhadap pertumbuhan bakteri S aureus termasuk dalam kategori sedang, dimana konsentrasi minimum getah J curcas dapat menghambat pertumbuhan bakteri S aureus yaitu 10 ppm.

Staphylococcus aureus (S aureus) is one of the pathogenic bacteria in Indonesia which became one of the causes of acne infection. Jatropha curcas Linn (J curcas) was identified as containing flavonoids, tannins, terpenoids, saponins and alkaloids which have antibacterial effects. This study aims to determine the compound content and activity of J curcas latex in inhibiting the growth of S aureus bacteria. J curcas latex is dried using an oven at 40°C for 7x24 hours to become a brownish amorphous crystalline. The dry latex obtained was screened for phytochemical using TLC test followed by TLC-Visualizer and then dried resin was analyzed by active compound using Ultra Performance Liquid Chromatography - Mass Spectrometry (UPLC-MS). The antibacterial activity test was performed by Kirby-bauer disc diffusion method uses 4 variations of concentration (25 ppm, 50 ppm, 75 ppm, and 100 ppm) and Minimum Inhibition Concentration (MIC) test was performed by dilution method (dilution series) or turbidimetry method. Positive control uses Amoxicillin 100 ppm while negative control uses water for injection (WFI). J curcas latex has a water-soluble amorphous crystalline form, ethanol and methanol. Based on the results of UPLC-MS analysis that has been done, J curcas latex contains pyridine compound which is a class of alkaloids and some other constituent compounds such as alcohol derivatives, carboxylic acid derivatives and cyclo (closed ring). The antibacterial activity of J curcas latex on bacterial growth S aureus belongs to the medium category, which the minimum concentration of J curcas can inhibit the growth of S aureus bacteria is 10 ppm.
