Abstract
INDONESIA:
Daun wungu (Graptophyllum pictum L. Griff.) merupakan tanaman obat yang layak dikembangkan sebagai salah satu tanaman obat unggulan asli Indonesia. Daun wungu mengandung alkaloid yang tidak beracun, glikosida, dan flavonoid yang kuat serta memiliki aktivitas anti-inflamasi, anti-plak, anti-oksidan, ant-bakteri, anti-diabetes dan sebagainya. Kebutuhan tanaman obat ini semakin meningkat sehingga perlu pengembangan dan budidaya. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi 2,4-D dan BAP serta kombinasinya dalam menginduksi kalus embriogenik daun wungu. Sehingga dapat dijadikan acuan dalam upaya perbanyakan bibit tanaman wungu secara in vitro.
Penelitian ini bersifat eksperimental, dengan menggunakan desain rancangan acak lengkap (RAL) 2 faktor, 25 kombinasi perlakuan dalam 3 ulangan. Faktor pertama adalah konsentrasi 2,4-D, yaitu 0 mg/l, 0.5 mg/l, 1 mg/l, 1.5 mg/l, dan 2 mg/l. Faktor kedua adalah konsentrasi BAP, yaitu 0 mg/l, 0.5 mg/l, 1 mg/l, 1.5 mg/l, dan 2 mg/l. Data yang diperoleh berupa data kuantitatif yang akan dianalisis menggunakan ANAVA dua jalur dengan α = 5% dan diuji lanjut menggunakan DMRT dengan taraf signifikansi 5%, serta uji regresi korelasi. Data kualitatif dianalisis menggunakan analisis deskriptif.
Hasil Penelitian menunjukkan bahwa konsentrasi 0.5 mg/l 2,4-D memberikan pengaruh nyata terhadap induksi kalus daun wungu, yaitu 16 HST, persentase tumbuh kalus pada eksplan 28,2% dengan berat basah kalus 0,04634 gr. Konsentrasi 0.5 mg/l BAP hanya berpengaruh nyata pada hari muncul kalus yaitu 18 HST. Konsentrasi kombinasi 1 mg/l 2,4-D + 0 mg/l BAP juga menunjukkan pengaruh nyata pada induksi kalus daun wungu, yaitu 12 HST, dan persentasi tumbuh kalus pada eksplan 73,3% dengan berat basah kalus 0,12307 gr. Morfologi dan Anatomi yang mencirikan kalus embriogenik yaitu pada perlakuan 0.5 mg/l 2,4-D + 0 mg/l BAP.

Wungu leaf (Graptophyllumpictum L. Griff.) is medicinal plants that are suitable to be developed as one of the superior original Indonesian medicinal plants. Wungu leaf contains non-toxic alkaloids, glycosides, strong flavonoids and it has anti-inflammatory, anti-plaque, anti-oxidant, anti-bacterial, anti-diabetic activities and so on. The need of medicinal plants is more increasing so it needs the development and cultivation. This study aims to know the influence of 2,4-D and BAP concentrations and their combinations in inducing embryogenic callus of wungu leaf. Therefore, it can be used as a reference in multiplicationof Wunguseeds in vitro.
This study was experimental by using Completely Randomized Design of 2 factors, 25 treatment combinations in 3 replications. The first factor is the concentration of 2,4-D, which is 0 mg / l, 0.5 mg / l, 1 mg / l, 1.5 mg / l, and 2 mg / l. The second factor is BAP concentration, which is 0 mg / l, 0.5 mg / l, 1 mg / l, 1.5 mg / l, and 2 mg / l. The data obtained was quantitative data that will be analyzed by using two-way ANAVA with α = 5% and further tested by using DMRT with a significance level of 5%, and correlation regression test. Qualitative data was analyzed by using descriptive analysis.
The results showed that the concentration of 0.5 mg / l 2,4-D gave real influence toward the induction of wungu leaf callus, which was 16 HST, with the percentage of callus growth on explants was 28.2% with callus wet weight 0.04634 gr. The concentration of 0.5 mg / l BAP only had real influence on the day the callus appeared which was 18 HST. The combination concentration of 1 mg / l 2,4-D + 0 mg / l BAP also showed real influence on the induction of wungu leaf callus, which was 12 HST, and the percentage of callus growth on explants was 73.3% with callus wet weight of 0.12307 gr. Morphology and anatomy that characterizes embryogenic callus, which is in the treatment of 0.5 mg / l 2,4-D + 0 mg / l BAP.
