Abstract
INDONESIA:
Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh 2,4-D, BAP dan interaksi 2,4-D dan BAP terhadap induksi kalus embriogenik Carica pubescens Lenne & K. Koch. Penelitian ini bersifat eksperimental, menggunakan rancangan acak lengkap (RAL) dengan 25 perlakuan dan 5 ulangan. Terdapat dua faktor dalam penelitan yaitu konsetrasi 2,4-D meliputi 0 mg/L, 2,5 mg/L, 5 mg/L, 7,5 mg/L, dan 10 mg/L dan konsentrasi BAP meliputi 0 mg/L, 0.25 mg/L, 0.5 mg/L, 0.75 mg/L dan 1 mg/L. Analisis dengan uji ANAVA Two Way α = 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5%. Hasil penelitian menunjukkan ada pengaruh pemberian zat pengatur tumbuh 2,4-D dan BAP. Konsentrasi 2,4-D yang paling efektif adalah 7,5 mg/L menginduksi kalus dengan hari muncul kalus 24,2 HST, berat basah kalus 0,96 g, persen luasan kalus pada eksplan 93,4%. Konsentrasi BAP yang paling efektif adalah konsentrasi 0.5 mg/L menginduksi kalus dengan hari muncul kalus 23,5 HST, berat basah kalus 0,96 g dan persen luasan kalus pada eksplan 95%. Interaksi 2,4-D dan BAP yang paling efektif adalah 7,5 mg/L 2,4-D dan 0,5 mg/L BAP dengan hari muncul kalus 19 HST, berat basah kalus 1,09 g dan persen luasan kalus pada eksplan 99%. Pengamatan morfologi dan anatomi menunjukkan ciri-ciri kalus embriogenik yang dibuktikan dengan kalus yang berwarna kekuningan dan bertekstur remah serta memiliki inti besar, mengandung butir pati, dan plastida.

This research aims to determine the effect of 2,4-D growth regulators, 2,4-D and BAP interactions on the induction of embryogenic callus Carica pubescens Lenne & K. Koch. This research is experimental, using a completely randomized design (CRD) with 25 treatments and 5 replications. There are two factors in research namely 2,4-D concentrations including 0 mg/L, 2,5 mg/L, 5 mg/L, 7,5 mg/L, and 10 mg/L and BAP concentrations include 0 mg/L, 0,25 mg/L, 0,5 mg/L, 0,75 mg/L and 1 mg/L. Analysis by testing ANAVA Two Way α = 5%. If there are significant differences then followed by Duncan Multiple Range Test (DMRT) with a significance level of 5%. Results of research shows that there is an influence of giving 2,4-D and BAP growth regulator. The most effective 2,4-D concentration is 7,5 mg/L inducing callus with days callus appeared 24,2 HST, callus wet weight 0,96 g, percent callus area in explants 93,4%. The most effective BAP concentration is the concentration of 0,5 mg/L induces callus with days of callus appearing 23,5 HST, callus wet weight 0.96 g and percent callus area at explants 95%. The most effective interactions of 2,4-D and BAP were 7,5 mg / L 2,4-D and 0,5 mg / L BAP with callus day 19 HST, callus wet weight 1,09 g and percent area callus in explants 99%. Morphological and anatomical observations show the characteristics of callus embryogenic as evidenced by yellowish and textured callus friable and have a large core, containing starch grains, and plastids.
