Abstract
INDONESIA:
Penelitian ini bertujuan mengetahui pengaruh zat pengatur tumbuh 2,4-D dan BAP terhadap induksi kalus metabolit sekunder ciplukan Physalis angulata. Bahan yang digunakan yaitu media MS, zat pengatur tumbuh, perkecambahan dari ciplukan yang diambil organ daun sebagai eksplan kalus. Kalus tersebut digunakan untuk memperbanyak metabolit sekunder ciplukan yaitu physalin yang digunakan sebagai antikanker dan dipercaya oleh masyarakat sebagai obat darah tinggi. Peningkatan kandungan metabolit sekunder ini diperoleh melalui kultur kalus kompak dengan penambahan zat pengatur tumbuh 2,4-D dan BAP sehingga produksinya meningkat. Penelitian ini bersifat eksperimental, menggunakan rancangan acak lengkap (RAL) dengan 25 perlakuan dan 3 ulangan. Perlakuan dalam penelitian ini ada dua faktor yaitu konsentrasi 2,4-D meliputi 0 mg/L, 0,5 mg/L, 1 mg/L, 1,5 mg/L, 2 mg/L dan konsentrasi BAP 0 mg/L, 0,25 mg/L, 0,75 mg/L dan 1 mg/L. Data dianalisis dengan uji ANAVA Two Way α= 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5% serta uji regresi kolerasi. Hasil dari penelitian kalus kompak menunjukkan adanya pengaruh konsentrasi 2,4-D dan BAP terhadap induksi kalus metabolit ciplukan Physalis angulata yang paling optimal pada kombinasi 0,5 mg/L 2,4-D dan 0,25 mg/L BAP hari muncul kalus hari ke-8 HST. Persentase 100% pada konsnetrasi 1 mg/L BAP dan berat kalus pada konsentrasi 1,5 mg/L 2,4-D dan 0,75 mg/L BAP berat kalus 0,5110 g.
ENGLISH:
This research is aimed at acknowledging the influence of growing regulatory substance 2,4-D and BAP towards the induction of ciplukan Physalis angulata secondary metabolism callus. The used media is MS, the growing regulatory substance, germination from ciplukan which is taken from the leaf organ as callus explant. This callus is used to multiply ciplukan’s secondary metabolism which is physalin used as anti cancer and believed by the society as high blood pressure medicine. The improvement of secondary metabolism substance is gotten through compact callus culture with the addition of growing regulatory substance 2,4-D and BAP thus the production is improving. This research is the kind of experimental, using the complete random design (RAL) with the 25 treatment and 3 repetation. The treatment in this research, there are two factors which is concentration 2,4-D covering 0 mg/L, 1 mg/L, 1.5 mg/L, 2 mg/L and concentration BAP 0 mg/L, 0,25 mg/L, 0,75 mg/L dan 1 mg/L. The data is analysed by ANAVA Two Way α= 5%. If there is significance difference thus followed by Duncan Multiple Range Test (DMRT) with the significant 5% also the regression correlation. The result of compact callus research shows that there is concentration influence 2,4-D and BAP tpwards callus induction metabolit ciplukan Physalis angulate which is most optimum for the combination 0,5 mg/L 2,4-D and 0,25 mg/L BAP days for callus in the days 8 HST. Percentage 100% for the concentration 1 mg/L BAP and the callus weight in the concentration 1.5 mg/L 2,4-D and 0,75 mg/L BAP callus weight 0,5110 g.
