Abstract
ABSTRAK
Penelitian ini bertujuan untuk mengetahui pengaruh metionin terhadap organogenesis somatik repetitif jeruk JC (Citrus limonia Osbeck.) dengan teknik TCL. Penelitian ini bersifat eksperimental, menggunakan rancangan acak lengkap (RAL) dengan 7 perlakuan dan 5 ulangan. Terdapat satu faktor dalam penelitian ini yaitu konsentrasi metionin meliputi 0 mg/L, 25 mg/L, 50 mg/L, 75 mg/L, 100 mg/L, 125 mg/L dan 150 mg/L. Analisis hasil pengamatan menggunakan ANAVA One way dengan α = 5 %, kemudian dilanjutkan dengan uji DMRT dengan taraf signifikan 5% apabila terdapat pengaruh yang nyata terhadap variabel pengamatan. Hasil penelitian menunjukkan adanya pengaruh pemberian metionin terhadap organogenesis somatik repetitif jeruk JC. Konsentrasi metionin yang paling efektif untuk organogenesis somatik repetitif jeruk JC adalah 75 mg/L yang mana mampu menginduksi persen eksplan berkalus sebesar 85,5%, hari muncul fase globular paling cepat yaitu 20,20 HST, jumlah tunas sebesar 9,35 tunas per eksplan, panjang tunas tertinggi yaitu 0,908 cm dan berat planlet 0,04 gram. Pengamatan morfologi pada planlet jeruk JC menunjukkan planlet yang ditanam dalam media dengan penambahan metionin sebesar 75 mg/L menghasilkan tunas yang paling banyak sehingga konsentrasi metionin paling optimal untuk menumbuhkan organogenesis somatik repetitif adalah 75 mg/L.
ABSTRACT
The purpose of the research to determine the effect of methionine on repetitive somatic organogenesis of citrus JC (Citrus limonia Osbeck.) with TCL techniques. This study was experimental, using a completely randomized design (CRD) with 7 treatments and 5 replications. There is one factor in this study, namely the concentration of methionine includes 0 mg / L, 25 mg / L, 50 mg / L, 75 mg / L, 100 mg / L, 125 mg / L and 150 mg / L. Analysis of observations using One way ANOVA with α = 5%, then continued with DMRT test with a significance level of 5% if there is a significant effect on the observation variable. The results of the study showed the influence of methionine administration on repetitive somatic organogenesis of JC oranges. The most effective methionine concentration for repetitive somatic organogenesis Orange JC is 75 mg / L which is capable of inducing percent of explants growing callus by 85.5%, the fastest globular phase of the day is 20.20 HST, the number of shoots is 9.35 shoots per explant, the highest shoot length is 0.908 cm and the weight of 0.04 gram plantlets. Morphological observations on JC citrus plantlets showed that plantlets planted in the media with the addition of 75 mg / L of methionine produced the most shoots so that the most optimal methionine concentration to grow repetitive somatic organogenesis was 75 mg / L.
