Abstract
INDONESIA:
Faktor penyebab utama penyakit jantung koroner adalah aterosklerosis yang ditandai dengan kadar malondialdehyde (MDA). MDA terbentuk dari peroksidasi lipid yang melibatkan aktivitas enzim siklooksigenase. Tujuan penelitian ini adalah 1) mengetahui pengaruh ekstrak kulit batang Cinnamomum burmanii dan daun Carica pubescens terhadap kadar MDA pembuluh koroner mencit secara in vivo; 2) mengetahui senyawa aktif yang berpotensi dalam menghambat enzim siklooksigenase secara in silico pada kulit batang Cinnamomum burmanii dan daun Carica pubescens. Penelitian ini bersifat deskriptif eksploratif (in silico) dan eksperimental dengan Rancangan Acak Lengkap (in vivo). Pengamatan secara in silico dilakukan dengan molecular docking sedangkan penelitian secara in vivo menggunakan spektrofotometri dengan 6 perlakuan dan 5 ulangan. Hewan coba yang digunakan adalah mencit jantan yang diinduksi HFD. Parameter yang diamati adalah kadar MDA pembuluh koroner. Data yang telah didapat kemudian dianalisis menggunakan One Way ANOVA. Hasil yang didapat dari uji in silico adalah tiga senyawa aktif yaitu cinnamate, cinnamaldehyde, dan ferulic acid dilihat dari nilai binding affinity yang rendah, HIA dan Pa yang tinggi serta memiliki asam amino residu yang sama dengan asam mefenamat. Hasil uji in vivo menunjukkan adanya pengaruh perlakuan P1, P2, dan P3 terhadap kadar MDA pembuluh koroner mencit. Kesimpulan dari penelitian ini adalah terdapat senyawa aktif yang berpotensi dalam mengurangi kadar MDA (in silico) dan ada pengaruh pemberian kombinasi ekstrak kulit batang kayu manis dan daun pepaya gunung terhadap kadar MDA pembuluh koroner mencit dislipidemia (in vivo).
ENGLISH:
The main cause of coronary heart disease is atherosclerosis which is characterized by levels of malondialdehyde (MDA). MDA is formed from lipid peroxidation which involves the activity of the cyclooxygenase enzyme. The purposes of the research are 1) to determine the influence of Extract on the Stem Bark of Cinnamomum burmanii and Carica pubescens leaves against MDA levels of mice coronary vessels through in vivo; 2) determine the active compounds to inhibit the cyclooxygenase enzyme through in silico on the steam bark of Cinnamomum burmanii and leaves of Carica pubescens. The research is descriptive exploratory (in silico) and experimental with Completely Randomized Design (in vivo). In silico observation was carried out by molecular docking, in vivo studies used spectrophotometry with 6 treatments and 5 replications. The experimental animals used male mice induced by HFD. The parameters were MDA levels of coronary vessels. The data were analyzed using One Way ANOVA. The results of in silico test were three active compounds, namely cinnamate, cinnamaldehyde, and ferulic acid that had been seen from low affinity binding values, high HIA and Pa and have the same residual amino acid withmefenamic acid. The results of in vivo test showed the effect of treatment of P1, P2, and P3 against MDA levels in the coronary vessels of mice. The conclusion of the research that there were active compounds that had the potential to reduce MDA (in silico) levels and there was an influence of the cinnamon bark extract gift and mountain (gunung) papaya leaves against MDA levels of dyslipidemia mice coronary vessels (in vivo).
