Abstract
INDONESIA:
Dislipidemia merupakan suatu kelainan metabolisme lipid yang ditandai dengan kenaikan kadar kolesterol total, trigliserida, LDL, dan penurunan HDL. Dislipidemia memiliki peranan penting dalam terbentuknya aterosklerosis yang merupakan penyebab penyakit jantung koroner. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak kulit batang kayu manis (Cinnamomum burmannii) dan daun pepaya gunung (Carica pubescens) terhadap kadar kolesterol total dan trigliserida serum darah mencit (Mus musculus). Penelitian ini bersifat deskriptif eksploratif dan eksperimental. Uji secara in silico menggunakan metode molecular docking antara reseptor HMG-KoA reduktase dengan ligan senyawa aktif pada kulit batang kayu manis dan daun papaya gunung. Parameter yang digunakan yaitu persentase HIA, nilai prediksi PASS dan nilai energi bebas (ΔG). Uji secara in vivo menggunakan RAL sebanyak 6 perlakuan dan 5 ulangan. Kelompok perlakuan yaitu N (normal), K- (HFD), K+ (HFD & atorvastatin), P1 (HFD & 300 mg/kgBB ekstrak kulit batang kayu manis), P2 (HFD & 150 mg/kgBB ekstrak kulit batang kayu manis & 150 mg/kgBB ekstrak daun papaya gunung), dan P3 (HFD & 300 mg/kgBB ekstrak daun papaya gunung). Induksi HFD dilakukan selama 56 hari dan pemberian dosis pengobatan dimulai pada hari ke-29 hingga hari ke-56 induksi HFD. Parameter yang digunakan yaitu kadar kolesterol total dan trigliserida pada serum darah. Analisis data menggunakan ANOVA dengan α=5%. Hasil melocular docking menunjukkan bahwa senyawa quercetin pada kulit batang kayu manis berpotensi dalam menghambat enzim HMG-KoA reduktase. Hasil analisis ANOVA menunjukkan bahwa perlakuan P1 (HFD & 300 mg/kgBB ekstrak kulit batang kayu manis) dapat menurunkan kadar kolesterol total. Selain itu P1, P2 dan P3 dapat menurunkan kadar trigliserida pada serum darah.

Dyslipidemia is a lipid metabolism disorder which is characterized by increased levels of total cholesterol, triglycerides, LDL, and decreased HDL. Dyslipidemia has an important role in the formation of atherosclerosis which is the cause of coronary heart disease. This study aims to determine the effect of Cinnamomum burmannii bark and Carica pubescens leaves extract on total cholesterol and triglycerides levels of mice blood serum. This research is descriptive exploratory and experimental study. In silico test using molecular docking method between HMG-CoA reductase receptor and active compound ligands of Cinnamomum burmannii bark and Carica pubescens leaves. The parameters used are HIA, PASS, and binding affinity (ΔG) score. In vivo tests using Completely Randomized Design (CRD) with 6 treatments and 5 replications. Treatment groups were N (normal), K- (HFD), K + (HFD & atorvastatin), P1 (HFD & 300 mg/kg extract of Cinnamomum burmannii), P2 (HFD & 150 mg/kg extract of Cinnamomum burmannii & 150 mg/kg mountain papa Carica pubescens leaves extract), and P3 (HFD & 300 mg/kg Carica pubescens leaves extract). HFD (High Fat Diet) induction was carried out for 56 days and treatment dose was started on the 29th to 56th day of HFD induction. The parameters used were total cholesterol and triglyceride levels in blood serum. Data was analyzed using ANOVA with α = 5%. Melocular docking results show that quercetin, a compound of Cinnamomum burmannii bark has the good potential to inhibit the HMG-CoA reductase enzyme. The ANOVA analysis results showed that P1 treatment (HFD + 300 mg/kg extract of Cinnamomum burmannii) significantly reduced total cholesterol levels. Furthermore, P1, P2 and P3 can significantly reduce triglyceride levels in blood serum.
