Abstract
INDONESIA:
Kantong plastik telah menjadi bagian yang tidak terpisahkan dari kehidupan masyarakat sehari-hari. Komposisi utama penyusun kantong plastik adalah salah satu jenis polimer etilen, yaitu Low Density Polyethylene (LDPE). Material ini diketahui memiliki ketahanan fisik yang tinggi dan sulit didegradasi di alam, sehingga berakhir mencemari lingkungan. Penelitian ini bertujuan untuk mengidentifikasi dan menentukan aktivitas degradasi jamur hasil isolasi dari tanah tempat pembuangan akhir (TPA) terhadap LDPE sebagai upaya untuk mengurangi jumlah sampah plastik LDPE di lingkungan.
Proses isolasi jamur dilakukan dalam media mineral salt (MSM) 1% glukosa dengan penambahan LDPE sebagai sumber karbon utama. Penentuan aktivitas degradasi isolat dilakukan setelah 40 hari masa inkubasi berdasarkan perhitungan weight loss. Ekstraksi DNA dilakukan dengan metode CTAB/NaCl (2%/1,5 M) yang dimodifikasi. Daerah ITS rDNA dipilih sebagai marker penanda untuk identifikasi molekuler dan diamplifikasi dengan metode Polymerase Chain Reaction (PCR) memakai pasangan primer ITS1/ITS4. Penentuan sekuen ITS rDNA isolat jamur dilakukan dengan metode sekuensing Sanger. Program komputer digunakan untuk melakukan analisis kekerabatan berdasarkan kedekatan sekuen query dengan database.
Isolat HJL M4LK1 yang didapatkan melalui proses isolasi diketahui memiliki aktivitas biodegradasi LDPE 13,33 ± 0,9428%. Ekstraksi DNA jamur dengan metode CTAB/NaCl menghasilkan isolat dengan rasio A260/A280 sebesar 1,83. Amplifikasi gen ITS dengan metode PCR pada suhu annealing 48oC mendapatkan DNA amplikon berukuran ~500 bp. Penentuan sekuen menggunakan metode dideoksi Sanger mendapatkan daerah ITS dengan panjang sekuen 426 bp. Urutan DNA hasil sekuensing dicontig dan dianalisis menggunakan program komputer. Beberapa strain jamur hasil analisis dipilih dan dibuat pohon filogenetik. Berdasarkan analisis kekerabatan diketahui isolat HJL M4LK1 memiliki kedekatan genetik dengan Geotrichum candidum voucher R. Kirschner strain RRK17, dengan persen similariti sebesar 100%.

Plastic bag has been such an inseparable material of human daily life. This material is mainly comprised of a type of ethylene polymer termed as low density polyethylene (LDPE). This polymer is known to have high physical resilience and not readily degraded in nature, ended up polluting the environment. The aim of this study is to identify as well as determine the degradation activity of fungi isolated from local dumping area (TPA) against LDPE substrat as an attempt to decrease the amount of LDPE plastic waste in environment.
Isolation process was carried out in mineral salt medium (MSM) with 1% glucose concentration and supplemented with LDPE as the main carbon source. The LDPE-degradation rate of fungal isolate was determined based on weight loss calculation after 40 day incubation period. DNA extraction was performed using modified CTAB/NaCl method. ITS rDNA was selected as a marker for molecular identification. Amplification process was carried out using Polymerase Chain Reaction (PCR) at annealing temperature of 48oC with ITS1/ITS4 as primer. ITS sequence determination was conducted through Sanger sequencing method. Bioinformatic analysis was performed using computational programs to do species identification by comparing sequence in query with those on database.
Through isolation process, isolate HJL M4LK1 was obtained and noted to have LDPE degradation activity of 13.33 ± 0.9428%. DNA extraction was carried out using CTAB/NaCl method with NaCl concentration of 1.5 M, yielding isolate of 1,83 purity based on A260/A280 ratio value. The amplification process was yielding DNA amplicon about ~500 bp in size. The ITS sequence obtained has 426 bp in size.. Phylogenetic tree construction of HJL M4LK1 showed the isolate has 100% similarity to Geotrichum candidum voucher R. Kirschner strain RRK17.
