Abstract
INDONESIA:
Hydrilla verticillata merupakan suku Hydrocharitaceae yang banyak tersebar di danau Ranu Pasuruan. Tumbuhan hydrilla mengandung senyawa metabolit sekunder yaitu steroid, alkaloid, saponin, tanin, flavonoid dan triterpenoid yang terbukti memiliki kemampuan sebagai obat. Penelitian ini bertujuan untuk mengetahui tingkat toksisitas fraksi n-heksana; kloroform dan n-butanol serta mengetahui golongan senyawa aktif yang memiliki toksisitas tertinggi terhadap Artemia salina Leach.
Tumbuhan hydrilla diekstrak dengan metode maserasi menggunakan pelarut metanol p.a. Ekstrak metanol dihidrolisis dengan larutan HCl 2 N kemudian dipartisi tidak bertingkat dengan variasi pelarut yaitu n-heksana, kloroform dan n-butanol. Masing-masing fraksi diuji toksisitasnya menggunakan metode Brine Shrimp Lethal Test dengan parameter nilai LC50. Identifikasi senyawa aktif dilakukan dengan uji fitokimia dilanjutkan dengan identifikasi menggunakan LC-MS/MS.
Hasil penelitian menunjukkan bahwa fraksi n-heksana memiliki toksisitas tertinggi terhadap larva udang Artemia salina Leach yaitu yang memiliki nilai LC50 terendah. Nilai LC50 masing-masing fraksi yaitu: fraksi n-heksana (30,8615 ppm), fraksi kloroform (34,6766 ppm), dan fraksi n-butanol (39,1573 ppm). Uji fitokimia pada ekstrak metanol menunjukkan adanya alkaloid, saponin, steroid dan tanin. Fraksi n-heksana menunjukkan adanya saponin dan steroid, fraksi kloroform mengandung alkaloid, saponin, triterpenoid dan steroid, sedangkan untuk fraksi n-butanol mengandung alkaloid, saponin, steroid dan tanin. Hasil pengujian steroid fraksi n-heksana dengan LC-MS/MS diperoleh yaitu 397, 383, 395, 369, dan 369 menunjukkan adanya steroid jenis ß-sitosterol, campesterol, stigmasterol, fukosterol dan kolesterol.

Hydrilla verticillata is a tribe of Hydrocharitaceae which is widely spread on Ranu Lake Pasuruan. Hydrilla plants contain secondary metabolites that is steroids, alkaloids, saponins, tannins, flavonoids and triterpenoids which has proven ability as a medicine. This study aims to determine the level of toxicity of the n-hexane fraction; chloroform and n-butanol and know the class of active compounds that have the highest toxicity to Artemia salina Leach.
Hydrilla plant is extracted by maceration method using methanol solvent p.a. Methanol extract was hydrolyzed with 2 N HCl solution then partitioned non-level with variations of solvents namely n-hexane, chloroform and n-butanol. Each of the fractions was tested for toxicity using the Brine Shrimp Lethal Test method with a parameter value of LC50. Identification of active compounds was carried out by phytochemical test followed by identification using LC-MS / MS.
The results showed that the n-hexane fraction had the highest toxicity to shrimp larvae Artemia salina Leach which had the lowest LC50 value. The LC50 values of each fraction were: n-hexane fraction (30.8615 ppm), chloroform fraction (34.6766 ppm), and n-butanol fraction (39.1573 ppm). Phytochemical test on methanol extract showed the presence of alkaloids, saponins, steroids and tannins. The n-hexane fraction showed the presence of saponins and steroids, chloroform fraction contained alkaloids, saponins, triterpenoids and steroids, whereas for the n-butanol fraction contained alkaloids, saponins, steroids and tannins. The results of steroid testing of n-hexane fraction with LC-MS / MS obtained were 397, 383, 395, 369, and 369 indicating the presence of ß-sitosterol, campesterol, stigmasterol, fukosterol and cholesterol steroids.
