Abstract
INDONESIA:
Bawang dayak merupakan tanaman endemik kalimantan Timur yang kini telah dibudidayakan pada berbagai lokasi di Indonesia. Bawang dayak memiliki berbagai macam manfaat salah satunya sebagai agen antikanker. Adanya perbedaan lokasi tumbuh mempengaruhi perbedaan kandungan metabolit pada bawang dayak. Penelitian ini bertujuan untuk mengetahui profil aktivitas antikanker terhadap sel kanker kolon (WiDr cell line) dan toksisitas terhadap sel normal (vero cell line) secara invitro terhadap ekstrak etanol 96% bawang dayak (Eleutherine palmifolia L.) dari beberapa lokasi yang berbeda di Indonesia. Pemisahan senyawa aktif umbi bawang dayak dilakukan dengan metode ultrasonik menggunakan pelarut etanol 96%. Masing- masing ekstrak yang berasal dari 6 provinsi yakni Kalimantan Timur, Kalimantan Tengah, Kalimantan Selatan, Jawa Timur, Jawa Barat, dan Jawa Tengah diuji aktivitas dan toksisitasnya terhadap WiDr cell line dan vero cell line menggunakan metode MTT assay. Hasil penelitian menunjukkan terdapat perbedaan uji aktivitas (IC50), toksisitas (CC50) dan nilai indek selektivitas. Hasil uji aktivitas pada Kalimantan Timur 133,2313 µg/mL, Kalimantan Selatan 269,796 µg/mL, Kalimantan Tengah 104,5183 µg/mL, Jawa Timur 86,981 µg/mL, Jawa Barat 159,0733 µg/mL, Jawa Tengah 272,5497 µg/mL, dan Cisplatin 47,16333. Hasil uji Toksisitas Kalimantan Timur 414,646 µg/mL, Kalimantan Selatan 446,4883 µg/mL, Kalimantan Tengah 528,768 µg/mL, Jawa Timur 476,8757 µg/mL, Jawa Barat 519,7043 µg/mL, Jawa Tengah 393,1613 µg/mL, dan Cisplatin 232,348 µg/mL. Dan nilai indeks selektivitas yaitu Kalimantan Timur 3,1, Kalimantan Selatan 1,7, Kalimantan Tengah 5,1, Jawa Timur 5,5, Jawa Barat 3,3, Jawa Tengah 1,4, Cisplatin 4,9.

Dayak Onion is an Endemic plant of East Kalimantan, which is now cultivated in various locations in Indonesia. One of the numerous advantages of Dayak onion is anticancer agent. The difference of locations where it grows affects the difference of metabolite contained in Dayak onion. This study aims to find out the in vitro anticancer activity profile on the colon cancer cell (WiDr cell line) and the in vitro toxicity on the normal cell (vero cell line) on the 96% ethanol extract of Dayak onion (Eleutherine palmifolia L.) from various locations in Indonesia. The separation of active compounds in Dayak onion’s root is conducted using ultrasonic method helped by 96% of ethanol as the solvent. The activity and toxicity of each extract taken from six provinces namely East Kalimantan, Central Kalimantan, South Kalimantan, East Java, West Java, and Central Java on WiDr cell line and vero cell line has been tested using MTT assay method. The results of the study indicate a difference in activity test (IC50), toxicity (CC50) dan the value of index selectivity. The result of toxicity test in East Kalimantan is 414,646 µg/mL, South Kalimantan is 446,4883 µg/mL, Central Kalimantan is 528,768 µg/mL, East Java is 476,8757 µg/mL, West Java is 519,7043 µg/mL, Central Java is 393,1613 µg/mL, and Cisplatin is 232,348 µg/mL. Moreover, the value of index selectivity of East Kalimantan is 3,1, South Kalimantan is 1,7, Central Kalimantan is 5,1, East Java is 5,5, West Java is 3,3, Central Java is 1,4, and Cisplatin is 4,9.
